The relationship between viral infection and autoimmune manifestations has been emerging as a significant focus of study, underscoring the intricate interplay infections immune system. The innate immune system relies on nucleic acid (NA) sensors to detect viral infections and trigger type I interferon (IFN-I) responses, which are crucial for antiviral defense. NA pattern recognition receptors RNA or DNA within various cellular compartments, initiating signaling pathways. Functionally distinct CD4+ helper T (Th) cell subsets, including Th1, Th2, Th17, and regulatory cells (Treg), play a pivotal role in the regulation of acquired immunity. Although key proteins involved Th differentiation have already been identified how proteogenomic landscape changes during activation remains unclear. To address this issue, we characterized signatures to each subsets by RNA sequencing liquid chromatography-assisted mass spectrometry, which enabled us simultaneously quantify more than 10,000 protein-coding transcripts 8,000 single-shot. The results indicated that receptor affected almost half transcript protein levels low correlative gene-specific manner, specific cytokine treatments modified profiles manner subsets: Th17 Tregs particularly exhibited unique compared other subsets.
