Abstract Diabetic foot ulceration (DFU) is a devastating complication of diabetes whose pathogenesis remains incompletely understood. Here, we profile 174,962 single cells from the foot, forearm, and peripheral blood mononuclear using single-cell RNA sequencing. Our analysis shows enrichment unique population fibroblasts overexpressing MMP1, MMP3, MMP11, HIF1A, CHI3L1 , TNFAIP6 increased M1 macrophage polarization in DFU patients with healing wounds. Further, spatially separated samples same patient spatial transcriptomics reveal preferential localization these associated toward wound bed as compared to edge or unwounded skin. Biofilms in chronic wounds, including diabetic foot ulcers, pressure and venous leg pose a major challenge to wound management. Herein, we report Janus-type antimicrobial dressing for eradication of biofilms wounds. The consists electrospun nanofiber membranes coupled with dissolvable microneedle arrays enable effective delivery database-designed peptide both inside outside biofilms. This exhibited high efficacy against broad spectrum resistant pathogens vitro. Importantly, such was able eradicate methicillin-resistant Staphylococcus aureus (MRSA) an ex vivo human skin infection model type II mouse after daily treatment without applying surgical debridement. Most importantly, the can also completely remove Pseudomonas aeruginosa MRSA, dual-species biofilm model. In addition, our computational simulations suggested that microneedles were more peptides than free drugs. Our results indicate dressings may provide management polymicrobial infections.

Abstract Delayed wound healing has a profound impact on patients, healthcare, and society. Platelet‐rich plasma (PRP) gel, as preparation for regenerative medicine, proven to be of clinical value in various treatments. Nevertheless, its weak mechanical properties consequent burst release effect have restricted application efficacy. Here, an engineered PRP dual‐network hydrogel (named DN gel) based sodium alginate is constructed through simple “one‐step” activation process. Its improved gelling property sustained growth factors may beneficial use. Evaluations rats indicate that gel promote terms rapid re‐epithelialization, up‐regulated factor levels early transitions the angiogenesis stages.
