Objective Irisin, released by muscles during exercise, was recently identified as a neuroprotective factor in mouse models of Alzheimer disease (AD). In cohort AD patients, we studied cerebrospinal fluid (CSF) and plasma irisin levels, sex interactions, correlations with biomarkers. Methods Correlations between CSF levels biomarkers (amyloid β 1‐42, hyperphosphorylated tau, total tau [t‐tau]) Clinical Dementia Rating Scale Sum Boxes (CDR‐SOB) were analyzed patients dementia (n = 82), mild cognitive impairment 44), subjective memory complaint 20) biologically characterized according to the recent amyloid/tau/neurodegeneration classification. Results reduced ( p < 0.0001), lower female patients. Moreover, correlated positively Aβ42 both r 0.379, 0.001) male 0.262, 0.05) negatively CDR‐SOB −0.234, only A negative trend also observed t‐tau all −0.144, 0.082) subgroup −0.189, 0.084). Interpretation The results highlight relationship pathology, especially females. Our findings offer perspectives toward use marker continuum. ANN NEUROL 2024;96:61–73

Irisin is a myokine that generated by cleavage of the membrane protein fibronectin type III domain-containing 5 (FNDC5) in response to physical exercise. Studies reveal irisin/FNDC5 has neuroprotective functions against Alzheimer's disease, most common form dementia elderly, improving cognitive function and reducing amyloid-β tau pathologies as well neuroinflammation cell culture or animal models disease. Although current ongoing studies on show promising results, further mechanistic are required clarify its potential meaningful therapeutic target for alleviating We recently found irisin treatment reduces pathology increasing activity/levels amyloid-β-degrading enzyme neprilysin secreted from astrocytes. Herein, we present an overview irisin/FNDC5's protective roles mechanisms

Sarcopenia, defined as the age-associated loss of muscle mass and increased fragility with age, is increasing worldwide. The condition often precedes development Alzheimer’s disease, thereby decreasing levels mobility physical activity in those affected. Indeed, has, some studies, been associated an risk disease other dementias. However, a detailed understanding interplay between both conditions not available needs to be thoroughly addressed. In following review, we focus on several genes, specifically APOE, BDNF, ACE, FTO, FNDC5, that have conditions. We also discuss epigenetic regulation each these genes along non-coding RNAs (ncRNAs) may role sarcopenic phenotypes. Finally, assert application systems biology will unravel relationship sarcopenia believe prevention older age reduce incidence debilitating cognitive decline.

Irisin is a protein resulting from proteolytic cleavage of fibronectin type III domain-containing 5 (FND5). The ability irisin to modulate adipocyte and control glucose metabolism in human metabolic diseases gave rise the hypothesis that could have pivotal role aging-related diseases. Although animal models, increased levels been positively associated with better health outcomes, humans, its remains controversial. To provide an overview main finding on older adults, comprehensive search was performed through MEDLINE-PubMed, Web Science, Scopus, Cochrane databases for studies conducted adults (≥60 years) published since 2012. After grouping analyzing articles based conclusion this narrative review included did not yield consistent evidence regarding association between or disease adults. Further are necessary clarify effective promoting longevity.
