Interleukin (IL)-1β is a pro-inflammatory cytokine whose levels are increased in the brains of Alzheimer’s disease (AD) patients. Despite role IL-1β pathology AD, fact that it expressed at very low makes challenging to measure, hence limiting its potential use as reliable biomarker. Moreover, being able accurately and reliably measure IL-1 β blood possible evaluate this biomarker inflammatory response AD. In study, we compared three quantification methodologies, Meso-Scale Discovery (MSD), both V-Plex S-Plex versions, Quanterix’s SIMOA (Single-Molecule Array), serum AD patients age-matched controls. These assays routinely used with high specificity sensitivity human patients, yet best our knowledge, no study has all techniques for their accuracy biomarkers. Our findings indicate two MSD can be control serum, but assay showed highest receiver operating characteristics (ROCs), an area under curve (AUC) 0.9532, which AUC values assay, 0.5660, 0.6632. Abstract Objectives To establish the reference intervals (RIs) of serum glial fibrillary acid protein (GFAP) measured by fully automated Lumipulse system. Methods The study population consisted 340 healthy individuals, including 251 blood donors and 89 outpatients, with a median age 56 years. Serum GFAP levels were G assay on platform G1200 (FUJIREBIO Inc., Tokyo, Japan). RIs (2.5th 97.5th percentiles) calculated for overall stratified sex groups. For population, males, females partitions, we employed nonparametric methods, while age-and-sex groups “robust” method, as recommended CLSI. Results RI in whole was 10.4–92.0 pg/mL. When considering differences, showed higher than males across all A positive correlation observed between (Spearman’s rho=0.55, p<0.001). Alzheimer's disease (AD) is a neurodegenerative disorder characterised by amyloid-β (Aβ), tau hyperphosphorylation and neurodegeneration. Blood-based biomarkers are emerging as minimally invasive tool for detection monitoring. This review depicts the relationships between modifiable lifestyle factors (nutrition, physical activity (PA), sleep, alcohol consumption, smoking, social isolation) plasma of AD: Aβ42, Aβ40, Aβ42/40, phosphorylated tau, total neurofilament light chain (NfL) glial fibrillary acidic protein. Limited evidence suggests that better nutrition associated with favourable AD biomarker profiles PA less NfL Aβ, whilst poor sleep elevated Aβ. However, lack data inconsistent findings highlight need further investigation to substantiate or refute these trends. Moreover, future research should include analysis on according gender, metabolic health APOE status. Considering growing emphasis preventing delaying dementia onset justified.
