ABSTRACT Central tolerance is an essential process that protects the mammalian immune system from developing autoimmune reactions by forming a self‐tolerant repertoire of T cells. The extent central depends on diversity self‐peptide–major histocompatibility complexes thymocytes encounter thymic antigen‐presenting cells (APCs). Decades research have demonstrated medullary epithelial (mTECs), unique type APC stromal origin, possess extraordinary capacity to produce and present thousands self‐peptides thymocytes. This ability facilitated various unconventional mechanisms, including AIRE‐regulated promiscuous gene expression, mimicry peripheral cell types, cooperative antigen transfer between different APCs. Recently, several studies reported mTECs other thymus‐resident also tonic inflammatory signaling, which shapes microenvironment expands presented inflammation‐associated self‐antigens (ISA). In this review, we focus interferons (IFNs), pro‐inflammatory molecules produced as mTECs.
