Traditional Chinese medicine Chaihu Shugan San (CSGS) is a classic herb prescription for improving depression, but its specific molecular mechanism has not been fully clarified. This study integrates network pharmacology and experimental validation to investigate CSGS's antidepressant effects, focusing on impact GC metabolism related pathways. In this research, the of CSGS in relation depression model induced by chronic unpredictable mild stress will be discussed. High-performance liquid tandem mass spectrometry was applied verification grown metabolites' economic vitality rat plasma prefrontal cortex. The revelation behavioral test results showed that administration improved symptoms significantly at end period, which 8 weeks. Network used assist verifying active ingredients affect glucocorticoid metabolic pathway exert effects. (GC) reducing corticosterone (CORT) levels increasing dehydrocorticosterone (11-DHCORT) 11-DHCORT/CORT ratio PFC. It regulated liver PFC downregulating synthase (11β-HSD1) upregulating enzymes (11β-HSD2). Additionally, restored signaling GR HSP-90α, FKBP51 HSP-70, alleviating inflammation inhibiting NF-κB P65 HAT expression. These particularly PFC, were stronger than those with fluoxetine. revealed targets multiple pathways including PI3K-Akt, FoxO, HIF-1, mTOR. indicate can improve depressive state rats regulating other as well downstream proteins.
