Abstract Regeneration and tissue turnover require new cell production positional information. Planarians are flatworms capable of regenerating all body parts using a population stem cells called neoblasts. The information required for patterning is primarily harbored by muscle cells, which also control contraction. Here we produce an in silico planarian matrisome use recent whole-animal single-cell-transcriptome data to determine that major source extracellular matrix (ECM). No other ECM-secreting, fibroblast-like type was detected. Instead, express core ECM components, including 19 collagen-encoding genes. Cell types are the basic building units of multicellular life, with extensive diversities. The evolution cell is a crucial layer comparative biology but thus far not comprehensively studied. Regenerative abilities vary dramatically across animals. Even amongst planarian flatworms, well-known for complete regeneration from tiny body fragments, some species have restricted while others are almost entirely incompetent. Here, we assemble a diverse live collection of 40 to probe the evolution head in group. Combining quantification species-specific head-regeneration with comprehensive transcriptome-based phylogeny reconstruction, show multiple independent transitions between robust whole-body and freshwater species. RNA-mediated genetic interference inhibition canonical Wnt signalling interference-sensitive bypassed all defects, suggesting that pathway is linked emergence defects. Our finding has roles reproductive system model Schmidtea mediterranea raises possibility trade-off egg-laying, asexual reproduction by fission/regeneration drives regenerative trait evolution.
