The process of aging is accompanied by a dynamic restructuring the immune response, phenomenon known as immunosenescence. This mini-review navigates through complex landscape age-associated changes, chronic inflammation, age-related autoimmune tendencies, and their potential links with immunopathology Long COVID. Immunosenescence serves an introductory departure point, elucidating alterations in cell profiles functional dynamics, changes T-cell receptor signaling, cytokine network dysregulation, compromised regulatory function. Subsequent scrutiny or “inflammaging,” highlights its roles susceptibilities mediator perturbations observed COVID patients. introduction epigenetic facets further amplifies interconnections. In this compact review, we consider interactions between immunosenescence, autoimmunity. We aim to explore multifaceted relationships that link these processes shed light on underlying mechanisms drive interconnectedness. With focus understanding immunological context aging, seek provide insights into how immunosenescence inflammation contribute emergence progression disorders elderly may serve for disturbances.

Since the emergence of SARS-CoV-2 virus in 2019, nearly 700 million COVID-19 cases and 7 deaths have been reported globally. Despite most individuals recovering within four weeks, Center for Disease Control (CDC) estimates that 7.5% to 41% develop post-acute infection syndrome (PAIS), known as 'Long COVID'. This review provides current statistics on Long COVID's prevalence, explores hypotheses concerning epidemiological factors, such age, gender, comorbidities, initial severity, vaccine interactions, delves into potential mechanisms, including immune responses, viral persistence, gut dysbiosis. Moreover, we conclude women, advanced non-vaccination, low socioeconomic status all appear be risk factors. The reasons these differences are still not fully understood likely involve a complex relationship between social, genetic, hormonal, other Furthermore, with seem more endure economic hardship due persistent symptoms. In summary, our findings further illustrate multifaceted nature COVID underscore importance understanding factors mechanisms needed effective therapeutic strategies interventions.

Postural orthostatic tachycardia syndrome (POTS) is a common accompaniment of variety chronic, inflammatory diseases, including long COVID, as are small, insoluble, 'fibrinaloid' microclots. We here develop the argument, with accompanying evidence, that fibrinaloid microclots, through their ability to block flow blood microcapillaries and thus cause tissue hypoxia, not simply correlated but in fact, by preceding it, may be chief intermediary POTS, which body's exaggerated 'physiological' response hypoxia. Similar reasoning accounts for symptoms bundled under term 'fatigue'. Amyloids known membrane disruptors, when targets nerve membranes, this can explain neurotoxicity hence autonomic nervous system dysfunction contributes POTS. Taken together view, we indicate microclots serve link POTS fatigue COVID manner at once both mechanistic explanatory. This has clear implications treatment such diseases.

Orthostatic intolerance is a broad term that represents spectrum of dysautonomic disorders, including postural orthostatic tachycardia syndrome (POTS) and hypotension (OH), as manifestations severe autonomic failure. While the etiology has not yet fully been uncovered, it associated with multiple underlying pathological processes, peripheral neuropathy, altered renin–aldosterone levels, hypovolemia, autoimmune processes. Studies have implicated adrenergic, cholinergic, angiotensin II type I autoantibodies in pathogenesis intolerance. Several case series demonstrated immunomodulation therapy resulted favorable outcomes, improving symptoms POTS OH. In this review, we highlight contemporary literature detailing association autoimmunity

Abstract Background Long COVID is a major public health burden causing diverse array of debilitating symptoms in tens millions patients globally. In spite this overwhelming disease prevalence and staggering cost, its severe impact on patients’ lives intense global research efforts, study the has proved challenging due to complexity. Genome-wide association studies (GWAS) have identified only four loci potentially associated with disease, although these results did not statistically replicate between studies. A previous combinatorial analysis total 73 genes that were highly two long cohorts predominantly (>91%) white European ancestry Sano GOLD population, we sought reproduce findings independent ancestrally more All Us (AoU) population. Methods We assessed reproducibility 5,343 signatures from original AoU Because very small population sizes provide limited power findings, initially tested whether observed significant enrichment are also positively correlated cohort after controlling for substructure. Results For case frequency greater than 5% AoU, consistently (77% - 83%, p < 0.01) cohort. These encompassed 92% study.
