Postural Orthostatic Tachycardia Syndrome (POTS) affects up to 1% of the US population, predominantly women, and is characterized by a complex, elusive etiology heterogeneous phenotypes. This review delves into intricate physiology POTS, decoding roles sinoatrial node, autonomic nervous system, fluid dynamics, interplay between immune endocrine systems. It further examines key contributing factors such as dysautonomia, thoracic hypovolemia, neuropathies, sympathetic denervation, autoimmune responses, associations with conditions small-fiber neuropathy mast cell activation syndrome. Given numerous mysteries surrounding we also cautiously bring attention node myocardial function, particularly in how heart responds stress despite exhibiting normal cardiac phenotype at rest. The potential genomic research elucidating underlying mechanisms POTS emphasized, suggesting this valuable approach that likely improve our understanding genetic underpinnings POTS. introduces tentative classification system for etiological which seeks capture condition's diverse aspects categorizing various acknowledging co-occurring conditions. classification, while aiming enhance optimize treatment targets, presented preliminary model needing study refinement. underscores ongoing need unravel complexities develop targeted therapies can patient outcomes.

Abstract Background: COVID-19 is a new multisectoral healthcare challenge. This study contributes to research on long COVID. It aims at understanding experiences, knowledge, attitudes and (information) needs that GPs have in relation COVID how these evolved since the beginning of pandemic. Methods: The used an exploratory qualitative design. investigated infection protection, infrastructure, framework conditions treatment patients during pandemic Austria through semistructured interviews. A total 30 interviews with different primary care settings (single practices, group centres) were conducted between February July 2022. For this study, questions relating analysed. The data analysed using content analysis software Atlas.ti. Results: first empirically long-COVID management by country Bismarck model place. All indicated having experience In cities, tended slightly better networks specialists, GPs, especially more rural places, would need much support. who already worked teams find easier. symptoms physicians described as Long-Covid correspond those international literature, but it unclear whether postCovid syndromes such PostExertinal-Malaise, autonomic dysfunction postural tachycardia syndrome or Mast-Cell-Overactivation-Syndrom (MCAS), cognitive are also recognized correctly classified they never mentioned. basically enormous challenge frustrating if does not yield significantly improved health. With problem, feel left alone. lacked information contact about specific health personnel points familiar postinfectious syndromes. Such facilities urgently needed. Conclusion: COVID-19 will continue preoccupy our systems for time come, variants without prevention strategies produce patients. Therefore, question when good support adequate pathways people be implemented.

Major determinants of the biological background or reserve, such as age, sex, comorbidities (diabetes, hypertension, obesity, etc.), and medications (e.g., anticoagulants), are known to affect outcome after traumatic brain injury (TBI). With unparalleled data richness coronavirus disease 2019 (COVID-19; ∼375,000 counting!) well chronic form, long-COVID, also called post-acute sequelae SARS-CoV-2 infection (PASC), publications (∼30,000 counting) covering virtually every aspect diseases, pathomechanisms, biomarkers, phases, symptomatology, etc., have provided a unique opportunity better understand appreciate holistic nature interconnectivity between organ systems, importance in modifying trajectories affecting outcomes. Such approach is badly needed TBI-induced conditions their totality. Here, I briefly review what about long-COVID/PASC, its underlying—suspected—pathologies, pathobiological changes induced by TBI, other words, TBI endophenotypes, discuss intersection long-COVID/PASC pathobiologies, how considering some factors person's inclusion mechanistic molecular biomarkers can help improve clinical management patients.

Postural orthostatic tachycardia syndrome (POTS) is characterized by exaggerated in the absence of hypotension. The pathophysiology POTS may involve hypovolemia, autonomic neuropathy, a hyperadrenergic state, and cardiovascular deconditioning, any which can co-occur same patient. Furthermore, there growing evidence role autoimmunity subset cases. In recent years, investigators have described an increased rate autoimmune comorbidities as evidenced finding several types neural receptor autoantibody non-specific marker patients with POTS. particular, association disease anti-G protein-coupled (GPCR) antibodies has frequently been noted. A previous study reported that autoantibodies to muscarinic AChRs play important persistent, gastrointestinal symptoms. To date, recognized one sequelae coronavirus 2019 (COVID-19) its frequency pathogenesis are still largely unknown. Multiple occur COVID-related, disorders, suggesting presence pathology these disorders. Herein, we review anti-GPCR disorders nervous system, particular POTS, provide new perspective for understanding POTS-related autoimmunity.

Abstract Postural orthostatic tachycardia syndrome (POTS) is a chronic, debilitating condition that characterized by an excessive increase in heart rate upon challenge. Before the COVID-19 pandemic, POTS affected 0.5% to 1% of U.S. population. Since incidence has risen sharply, adding estimated 6-7 million new cases Despite its importance, there currently no reliable biomarker for POTS, leading significant diagnostic delays. A major hurdle identifying biomarkers heterogeneous nature syndrome. To address this, we focused on homogeneous subgroup post-COVID-19 (PC-POTS) patients. We conducted quantitative proteomics sera from 9 PC-POTS patients and healthy controls, 31 proteins with significantly different abundances Most elevated were linked actin filaments or immune functions/inflammation. Weighted Gene Co-Expression Network Analysis revealed module 7 (M7) correlated strongly diagnosis related traits. The key M7 included MTPN, TAGLN2, ADP-ribosylation factor 1, PDLIM1, PPIA, CNN2, LGALSL, TXN, TLN1, TUBA4A, IL4, TREML1, GP1BA, and, all highly these Cell-type enrichment analysis was associated neuronal cells. main pathways identified integrin signaling pathway, blood coagulation, glycolysis. These findings suggest could serve as PC-POTS. This study uses identify potential differentiate establishing foundation further research validation.
