This study aimed to investigate age-specific alterations in functional connectivity after sleep deprivation (SD) and decode brain changes from neurocognitive transcriptomic perspectives. Here, we examined global regional graph measures, particularly network strength (RNS), 41 young participants 36 older with normal 3 h of SD. Additionally, by utilizing cognitive probabilistic maps Neurosynth gene expression data the Allen Human Brain Atlas, applied partial least-squares regression analysis identify transcriptional correlates these RNS changes. After SD, exhibited decreased default mode (DMN) dorsal attention network, increased visual network. Young also showed DMN, notably left inferior parietal lobe, dorsolateral prefrontal cortex, posterior cingulate cortex. In participants, SD-induced significantly correlated processes such as "attention," "cognitive control," "working memory," while they "learning," "focus," "decision." Gene-category enrichment indicated that specific genes related signal transduction, ion channels, immune signaling might influence SD pathophysiology affecting participants. elucidates shared associated providing a molecular basis for understanding underlying pathophysiology.

ABSTRACT This study integrated neuroimaging and neurochemistry data to explore brain mechanisms in chronic insomnia disorder (CID) the neuromodulatory effects of acupuncture. We analyzed a cross‐sectional arterial spin labeling (ASL) dataset ( N = 197) CID patients healthy controls identify cerebral blood flow (CBF) changes. Additionally, longitudinal ASL 44) examined CBF changes after 4‐week acupuncture treatment or on waitlist. then assessed impact 19 neurotransmitter receptors/transporters these alterations. In comparisons, exhibited increased cortical areas decreased subcortical regions, correlating with severity. enhanced alleviated symptoms, not observed waitlist group. The left putamen was identified as an overlapping region involved both CID‐related post‐treatment Moreover, patterns induced by negatively correlated abnormal patients, were significantly associated GABAa dopamine‐D1 receptor densities. decrease could potentially serve neural biomarker for CID, while may alleviate symptoms increasing this region, through modulation D1 expressions.

Chronic insomnia disorder (CID) is a multidimensional disease that may influence various levels of brain organization, spanning the macroscopic structural connectome to microscopic gene expression. However, connection between genomic variations and morphological alterations in CID remains unclear. Here, we investigated changes patients at whole-brain level whether these link transcriptional characteristics. Brain data from 104 102 matched healthy controls (HC) were acquired examine cortical using morphometric similarity (MS) analysis. Partial least squares (PLS) regression transcriptome Allen Human Atlas used extract genomes related MS changes. Gene-category enrichment analysis (GCEA) was identify potential molecular mechanisms behind observed We found exhibited reductions parietal limbic regions, along with enhancements temporal frontal regions compared HCs (p
