Since the emergence of SARS-CoV-2 Omicron BA.1 and BA.2, several sublineages have emerged, supplanting their predecessors. Here we compared neutralization BA.1, BA.4 BA.5 by human sera collected from individuals who were infected with ancestral B.1 (D614G) strain, vaccinated (3 doses) or breakthrough infection pre-Omicron strains (Gamma Delta). All exhibited extensive escape all when to strain Delta, albeit different levels depending on origin sera. Convalescent unable neutralize partly neutralized BA.5. Vaccinee but evaded neutralizing antibodies (NAb). Some infections (BTI) non-neutralizing. Neutralizing BTI had similar ability against sublineages. Despite anti-Spike anti-Receptor Binding Domain (RBD) in groups, highest cross-neutralizing convalescent least neutralizing. Antibody avidity inferred NT50:antibody titer ratio was patients, underscoring qualitative differences elicited vaccination. Together, these findings highlight importance vaccination trigger highly cross-reactive that phylogenetically antigenically distant strains, suggest immune imprinting first generation vaccines may restrict, not abolish, cross-neutralization.
