Abstract Neuropeptides are ubiquitous intercellular signaling molecules in the CNS and play diverse roles modulating physiological functions by acting on specific G-protein coupled receptors (GPCRs). Among them, elevenin system is now believed to be present primarily protostomes. Although was first identified from L11 neuron of abdominal ganglion mollusc Aplysia californica , no have been described nor any other molluscs. Here, using two annelid Platynereis dumerilii we found three putative GPCRs . We cloned tentatively named them apElevR1, apElevR2, apElevR3. Using an inositol monophosphate (IP1) accumulation assay, demonstrated that with disulfide bond activated low EC50 values (ranging 1.2 25 nM), supporting they true for elevenin. In contrast, without could not activate receptors, indicating required receptor activity. alanine substitution individual conserved residues than cysteines, showed these appear critical activity, different had sensitivities single residue substitution. Finally, examined those outside ring removing also appeared important Thus, our study provides basis further its

Animals' survival is dependent on their abilities to adapt the changing environment by adjusting behaviours, which related ubiquitous learning behaviour, nonassociative learning. Thus mimicking indispensable behaviour in organisms based electronic devices vital better achieve artificial neural networks and neuromorphic computing. Here a three terminal device consisting of an oxide-based memristor NMOS transistor proposed. The with gradual conductance tuning inherently functions as synapse between sensor neurons motor presents adjustable synaptic plasticity, while attached utilized mimic modulatory effect neuromodulator released inter neurons. Such memristor-based multi-terminal allows practical implementation significant single device. In this study, experience-induced modification both habituation sensitization, was successfully achieved. dependence behavioural response strength interval presented stimuli also discussed. offers feasible experimental advantages for further study systems devices.

The precise nature of the engram, physical substrate memory, remains uncertain. Here, it is reported that RNA extracted from central nervous system Aplysia given long-term sensitization (LTS) training induced when injected into untrained animals; furthermore, RNA-induced sensitization, like training-induced required DNA methylation. In cellular experiments, treatment with trained animals was found to increase excitability in sensory neurons, but not motor dissociated naïve animals. Thus, behavioral, and a subset cellular, modifications characteristic form nonassociative memory (LTM) can be transferred by RNA. These results indicate sufficient generate an engram for LTS are consistent hypothesis epigenetic changes underlie storage .

Important insights into the selection pressures and core molecular modules contributing to evolution of pain-related processes have come from studies nociceptive systems in several molluscan arthropod species. These phyla, chordates that include humans, last shared a common ancestor approximately 550 million years ago. Since then, animals these phyla continued be subject traumatic injury, often predators, which has led similar adaptive behaviors (e.g., withdrawal, escape, recuperative behavior) physiological responses injury each group. Comparisons across taxa provide clues about contributions convergent conservation ancient mechanisms general functions. Primary nociceptors been investigated extensively few species, with long-lasting sensitization gastropod, Aplysia, insect, Drosophila, being especially fruitful. In as model for aversive memory hyperalgesia. Neuromodulator-induced, activity-dependent, axotomy-induced plasticity defined synapses, cell bodies, axons Aplysia primary nociceptors. Studies Drosophila larvae revealed numerous contributors interacting cells. Interestingly, examined thus far are largely different, but both sets overlap those mammalian pathways. contrast results moth (Manduca) disclosed central hyperactivity no obvious peripheral responses. Squid (Doryteuthis) show injury-induced manifested behavioral hypersensitivity tactile visual stimuli, spontaneous activity nociceptor terminals. Temporary blockade during subsequently increased mortality when injured squid were exposed fish providing first demonstration any animal adaptiveness sensitization. Immediate noxious stimulation also behaviorally physiologically snail (Helix), octopus (Adopus), crayfish (Astacus), hermit crab (Pagurus), shore (Hemigrapsus). Molluscs arthropods suppress responses, whether opioid play antinociceptive roles is uncertain.

This review describes the advantages of adopting a molluscan complementary model, freshwater snail Lymnaea stagnalis, to study neural basis learning and memory in appetitive avoidance classical conditioning; as well operant conditioning its aerial respiratory escape behaviour. We firstly explored 'what we can teach Lymnaea' by discussing variety sensitive, solid, easily reproducible simple behavioural tests that have been used uncover abilities this model system. Answering question will allow us open new frontiers neuroscience research enhance our understanding how nervous system mediates memory. In fact, from translational perspective, help understand transformation pathways output sensory coding more complex systems like mammalian brain. Moving on second question: us?', it is now known shares important associative characteristics with vertebrates, including stimulus generalization, generalization extinction discriminative learning, opening possibility use snails animal models for research.
