Cellular senescence plays a crucial role in skin aging, with senescent dermal fibroblasts contributing to reduced elasticity and increased inflammation. This study investigated the potential of Ganoderma lucidum (Reishi) ethanol extract modulate phenotype human fibroblasts. Reishi powder two different vendors was used. Ganoderma lucidum (Leyss. ex Fr.) Karst, commonly known as Lingzhi, has long been employed in traditional Chinese medicine for its medicinal properties, particularly alleviating respiratory issues like cough and asthma. Recognized both a therapeutic agent an edible supplement, Lingzhi is celebrated health-promoting benefits. Abstract Background: In the in-vitro expansion of mesenchymal stem cells (MSCs), replicative or stress-induced senescence poses a significant challenge, leading to loss their cellular properties and therapeutic functions. Currently, there is lack efficient strategies address this issue. Ganoderma acid A (GAA), a triterpenoid compound from lucidum, has gained attention for its anti-tumor properties. Herein, we hypothesized that GAA may enhance cisplatin's (DDP) anticancer effect in gallbladder cancer (GBC) cells by promoting DNA damage response, particularly through upregulation of markers such as γH2AX, p-ATM, p-ATR, and p-p53, reducing cell stemness downregulating like SOX2, Oct4, NANOG. The human GBC line GBC-SD epithelial HGBEC were cultured RPMI-1640 DMEM/F12 media with 10% fetal bovine serum. Cells treated 2 µM DDP 60 24 h.
