This study investigated the association between NAFLD and liver fibrosis ratio of gamma-glutamyl transferase to high-density lipoprotein cholesterol (GGT/HDL-C). In this cross-sectional study, we included 4764 subjects who participated in National Health Nutrition Examination Survey (NHANES) during 2017–2018. Adjusted multivariate logistic regression analysis was utilized evaluate relationships GGT/HDL-C levels NAFLD, fatty degree, fibrosis. The non-linear link examined using generalized additive models. There a risk all models demonstrated strong relationship fibrosis, degree hepatic steatosis, prevalence NAFLD. Subgroup analyses revealed significant correlation among Mexican Americans young people 20–40 age range. receiver operating characteristic (ROC) showed that more accurate predictor than GGT or HDL-C alone. U.S. population, an increased severity are independently correlated with elevated ratio.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has become one of the most prevalent chronic diseases worldwide. The serum uric acid-to-high-density lipoprotein cholesterol ratio (UHR) been recognized as a novel marker for metabolic diseases, including MASLD. However, all previous studies were performed in adults. To explore relationship between UHR and MASLD Chinese children with obesity. A retrospective study was conducted 1284 obese hospitalized at Beijing Children's Hospital January 2016 December 2022. Logistic regression analysis restricted cubic splines to assess association odds receiver operator characteristic (ROC) curve used estimate diagnostic value prevalence high, which reached 61.76% levels higher than those non-MASLD both genders. After dividing individuals into three groups according tertiles UHR, rate increased progressively from tertile 1 3 (34.11% vs. 70.56% 80.61%). showed that significantly associated risk, independent confounding factors such age, gender, body mass index (BMI), fasting blood glucose (FBG), alanine aminotransferase (ALT), aspartate (AST), gamma-glutamyl transferase (GGT), low-density (LDL-C), triglycerides (TG), creatinine (Cr). non-linear demonstrated approximately 300 900 suggested saturation effect risk. ROC indicated might serve predictive diagnosing children. In obesity, is useful predictor onset.

Ophiopogon japonicus (Thunb.) Ker Gawl., is a traditional Chinese medicine (TCM) with history of thousands years. O. polysaccharides (OJPs), as one the main active ingredient, are kind representative pharmacological bioactive macromolecules and mainly composed Glc Fru molecular weight between 2.48 325 kDa. OJPs have variety biological activities, such hypoglycemic, cardioprotective, immunomodulatory, improvement obesity, renal protective activity. In this review, extraction, purification structural characterization were systematically reviewed, its function, mechanism structure–activity relationship analyzed. Additionally, review summarized found that modifications liposome modification, sulfation polyethylene glycol modification improved activity bioavailability OJPs. Overall, work will help to better understand provide theoretical their further development utilization in field health food.

ABSTRACT Objectives Metabolic dysfunction‐associated steatohepatitis (MASH) is a significant risk factor for cirrhosis and hepatocellular carcinoma, which there currently no effective treatment. This study aimed to investigate the regulatory mechanism between endoplasmic reticulum stress (ER stress) pyroptosis in liver under context of MASH. Methods Results Pyroptosis was examined both vivo vitro ER models. The expression levels nucleotide‐binding oligomerisation domain‐like receptor protein 3 (NLRP3), gasdermin D (GSDMD), caspase‐1, IL‐1β, IL‐18 tended increase, “ASC specks” colocalised with swollen living cells. However, pyroptotic model, increased not observed. Moreover, overexpression inositol‐requiring enzyme 1α (IRE‐1α), one main sensors, led increases NLRP3 GSDMD. after IRE‐1α blocked by chemical inhibitors or siRNAs, also abrogated. These data showed that regulated through IRE‐1α. Furthermore, immunoprecipitation results clearly indicated GSDMD efficiently bound when stimulated. In MASH specifically inhibited pharmacological genetic methods, improved pathology alleviating pyroptosis. patients MASH, markers including IRE‐1α, glucose‐regulated 78, GSDMD/GSDMD‐N, p20, NLRP3, are highly expressed liver. Conclusions revealed may regulate IRE‐1α‐GSDMD pathway, accelerates progression findings offer new insights treatment

Tissue fibrosis results from a dysregulated and chronic wound healing response accompanied by inflammation angiogenesis. Regardless of the affected organ, shares following common hallmarks: recruitment immune cells, fibroblast activation/proliferation, excessive extracellular matrix deposition. Chemokines play pivotal role in initiating advancing these fibrotic processes. CCL24 (eotaxin-2) is chemokine secreted cells epithelial which promotes trafficking activation profibrotic through CCR3 receptor binding. Higher levels were found tissue sera patients with fibro-inflammatory diseases, including primary sclerosing cholangitis (PSC), systemic sclerosis (SSc), metabolic dysfunction-associated steatohepatitis (MASH). This review delves into intricate highlighting its impact on fibrotic, immune, vascular pathways. We focus preclinical clinical evidence supporting therapeutic potential blocking diseases that involve fibrosis.

Non-alcoholic fatty liver disease (NAFLD) is a global cause of dysfunction. This spectrum hepatic disorders can progress to severe conditions, such as non-alcoholic steatohepatitis (NASH) and cirrhosis, due oxidative stress sustained cellular injury. With limited pharmacological options, glutathione (GSH), key antioxidant, has shown promising potential in reducing stress, maintaining redox balance, improving function. literature review examines studies from 2014–2024 exploring GSH therapy NAFLD patients. Eligible assessed the primary intervention for human subjects, reporting outcomes function or markers. Randomized clinical trials (RCTs) were eligible, while combination included if GSH’s effect could be isolated. Exclusions applied non-NAFLD studies, animal/in vitro models, non-GSH antioxidant interventions. Analysis three (totaling 109 participants) demonstrated consistent improvements alanine transaminase (ALT) levels reductions markers like 8-hydroxy-2-deoxyguanosine (8-OHdG). However, small sample sizes inconsistent protocols limit generalizability. Further large-scale RCTs are required confirm efficacy, determine optimal dosing, assess long-term effects. highlights novel therapeutic strategy emphasizing need further refine its application.

Noninvasive tests (NITs) are used to risk-stratify metabolic dysfunction-associated steatotic liver disease. The aim was survey global patterns of real-world use NITs. A 38-item designed by the Global NASH Council. Providers were asked about risks for advanced fibrosis, which NITs (cutoff values) they disease, monitor progression, and professional guidelines follow. total 321 participants from 43 countries completed (54% hepatologists, 28% gastroenterologists, 18% other). Of respondents, 85% would patients with type 2 diabetes, obesity (82%), or abnormal enzymes (73%). Among rule out significant transient elastography (TE) fibrosis-4 (FIB-4) most used, followed NAFLD Fibrosis Score, Enhanced Liver Fibrosis, magnetic resonance elastography. cutoffs ruling fibrosis varied considerably between practices guidelines, only 50% using TE <8 kPa, 65% FIB-4 <1.30 age <65, 41% <2.00 ≥65. Similar variability found in where thresholds ≥2.67 ≥10 kPa 20% 17%, respectively. To establish 48% while 23% consider 1 NIT, 17% confirm biopsy. >75% monitor, 66% (intermediate high risk) annually. Finally, follow guideline recommendations regarding In clinical practice, there is NIT their thresholds. Additionally, suboptimal adherence societies' guidelines.
