Pancreatic cancer (PC) is a lethal type of for which effective therapies are limited. Long non‑coding RNAs (lncRNAs) represent critical regulator category, mediating the tumorigenesis and development various tumor types, including PC. However, expression patterns functions numerous lncRNAs in PC remain poorly understood. In present study, linc01614 was identified as PC‑related lncRNA. notably upregulated tissues cell lines associated with poor disease‑free survival patients according to analysis The Cancer Genome Atlas‑derived datasets. Functionally, knockdown suppressed proliferation, migration invasion in vitro, inhibited proliferation vitro vivo. Mechanistically, overexpression stabilized level β‑catenin protein hyperactivate WNT/β‑catenin signaling pathway cells. Further analyses revealed that bound GSK‑3β perturbed interaction between AXIN1, thereby preventing formation degradation complex reducing β‑catenin. summary, findings reveal may function an oncogene promote progression thus be considered potential therapeutic target future.

Never-in-mitosis gene A-related-kinase-2 (NEK2) plays a pivotal role in malignant progression and chemotherapy sensitivity. This study aimed to elucidate the of NEK2 colorectal cancer (CRC) its potential contribution 5-fluorouracil (5‑FU) resistance mechanisms. Quantitative real-time PCR (qRT‑PCR), western blotting, immunohistochemical (IHC) staining were used assess expression CRC tissues cells. The effects 5‑FU on proliferation, apoptosis, migration, invasion cells investigated via Cell Counting Kit-8 (CCK-8), colony formation, flow cytometry, wound healing, transwell assays, respectively. Methyl 3-(4-methylphenylsulfonamido) benzoate (MSAB) was as Wnt/beta (β)-catenin pathway inhibitor this study. significantly upregulated compared normal controls. High correlated with advanced tumor-node-metastasis (TNM) stage, lymph node metastasis, distant poor tumor prognosis. overexpression promoted inhibited cytotoxic effect 5-FU nuclear accumulation β-catenin activated Wnt/β-catenin signaling pathway. MSAB reversed stimulatory upregulation proliferation In summary, promotes cell survival decreases sensitivity by activating Consequently, holds promise therapeutic target for management.

Osteopontin (OPN) is a multifunctional phosphorylated glycoprotein that expressed at significantly elevated levels in various cancers.OPN overexpression closely associated with the development of cancer progression such as proliferation, metastasis, angiogenesis, apoptosis resistance, drug and immunosuppression, may also be an independent prognostic biomarker for variety cancers. This review broadly summarizes mechanisms regulate expression downstream oncogenic molecules after OPN binds to integrin receptors or CD44 receptors, which involve complex intracellular "signaling traffic network" (including key kinases, signaling pathways, transcription factors). In addition, we value OPN, synergistic female breast, non-small cell lung, prostate, colorectal, gastric, hepatocellular carcinomas. The tissues blood vary due differences study subjects detection methods, this aspect requires further systematization view applying clinical applications.Importantly, based on fact effect correlates above-mentioned molecules, work provide some help combination therapy targeting molecules.

B-cell CLL/lymphoma 9 (BCL9) is considered a key developmental regulator and well-established oncogenic driver in multiple cancer types, mainly through potentiating the Wnt/β-catenin signaling. However, increasing evidences indicate that BCL9 also plays Wnt-independent roles. Herein, we summarized updates of canonical non-canonical functions cellular, physiological, or pathological processes. Moreover, concluded targeted inhibitors disrupt interaction β-catenin with reported recently.
