An allergen exposure chamber (AEC) is a specialized medical facility designed to expose individuals allergens at precise and consistent concentrations within controlled environment. This study aimed correlate the assessment of clinical endpoints in patients with allergic rhinitis sensitized timothy grass pollen (Phleum pratense) by comparing three different methods: AEC, nasal challenge (NAC), symptoms during natural season. Fifteen subjects twelve healthy controls were evaluated ALLEC AEC; measured subjective objective methods, including total symptom score (TNSS), acoustic rhinometry (AcR), peak inspiratory flow (PNIF), discharge amount. For in-field evaluation, combined medication (EAACI-CSMS), as proposed European Academy Allergy Clinical Immunology (EAACI), was used, which assessed for 14 days The AcR, PNIF, TNSS used NAC assessment. Both AEC NAC, all developed optimal pollen/allergen concentration across endpoints. Using Spearman's rank correlation coefficient (p < 0.05), analysis revealed significant correlations between 120 min challenge, well after 15 EAACI-CSMS in-field. stronger than those showed weaker or no correlations. indicates that correlates better assessments NAC. significantly correspond efficacy (EAACI-CSMS exposure). effect not outcomes measured. methods might be considered an alternative traditional exposure.

Allergic rhinitis (AR) is global health concern with an increasing prevalence. Among them, pollen-induced AR (PIAR) exhibits more severe and intense symptoms, decreased quality of life, prominent local inflammation, thus challenging to control. Due the difficulties in disease control, recent years, number treatment methods, including pharmacotherapy, allergen-specific immunotherapy, newly developed biologics, have focused on PIAR. It has been shown that pollen exposure a significant impact symptoms PIAR efficacy intervention. From this perspective, clinical trials for need take full account exposure, especially when assessing efficacy. This review summarized effect PIAR, immune responses, clinic visits. Current definitions season (PS) peak (PPS) are discussed. Based previous PIAR-related studies available recommendations trial design, detailed protocols which fully considered provided. Pollen With considering design future may be objective better harmonized and, therefore, comparable.

Allergic diseases triggered by airborne allergens such as allergic rhinitis and conjunctivitis are increasingly prevalent, posing significant challenges for both patients healthcare systems. Assessing the efficacy of allergen immunotherapy other anti-allergic treatments requires precise reproducible methods. Allergen exposure chambers (AECs) have emerged advanced tools evaluating clinical outcomes, offering controlled conditions that address many limitations traditional field-based studies. This review explores advantages AECs in allergy management, emphasizing their role providing standardized research routine assessments. deliver consistent data comparable to nasal challenge natural exposure, making them a valuable addition diagnosis treatment effectiveness diseases. Although they well suited early-stage trials, further standardization validation needed gain broader acceptance pivotal phase III Future should focus on refining AEC protocols integrating into regulatory frameworks, ensuring advancement therapeutic approaches
