Oxidative stress plays a role in various pathophysiological diseases, including neurogenerative such as Alzheimer′s disease (AD), which is the most prevalent neuro-pathology aging population. has been reported to be one of earliest pathological alterations AD. Additionally, it was demonstrated that older adults, there loss free radical scavenging ability. The Nrf2 transcription factor key regulator antioxidant defense systems, but, with aging, both amount and transcriptional activity decrease. With available treatments for AD being poorly effective, reinforcing systems via pathway may way prevent treat To highlight predominant signaling defending against oxidative and, therefore, neurotoxicity, we present an overview natural compounds exert their own neuroprotective roles through activation pathway. This review opportunity promote holistic approach treatment need further refine development new potential Nrf2-targeting drugs.

Ischemic stroke (IS), a major cause of death and disability globally, requires innovative therapeutic approaches due to its complex pathology. Nature medicine (NM) offers promising treatments through bioactive compounds, which target the multifaceted nature stroke-induced damage. However, clinical application NM is limited by challenges in bioavailability specificity. This review article presents an advanced perspective on integrating nanotechnology with create potent nanodelivery systems for ischemic treatment. We highlight pathological underpinnings stroke, including oxidative stress, inflammation, apoptosis, discuss how compounds offer targeted mitigation strategies. By incorporating platforms, such as liposomes nanoparticles, these -based can achieve enhanced targeting, solubility, controlled release, significantly improving outcomes while reducing side effects. Despite developments, translation nano-enhanced into practice faces obstacles, manufacturing scalability, regulatory approval, safety evaluations. emphasizes potential combining advance therapy, calling integrated research efforts overcome existing barriers fully realize benefits this approach.

Aims Effective intervention for mild cognitive impairment (MCI) is key preventing dementia. As a neuroprotective agent, butylphthalide has the potential to treat MCI due Alzheimer disease (AD). However, pharmacological mechanism of from brain network perspective not clear. Therefore, we aimed investigate multimodal changes associated with treatment in AD. Methods A total 270 patients AD received either or placebo at ratio 1:1 1 year. was defined as decrease Alzheimer's Disease Assessment Scale‐Cognitive Subscale (ADAS‐cog) > 2.5. Brain networks were constructed using T1‐magnetic resonance imaging and fluorodeoxyglucose positron emission tomography. support vector machine applied develop predictive models. Results Both (drug vs. placebo)‐time interactions efficacy (effective ineffective)‐time detected on some overlapping structural metrics. Simple effects analyses revealed significantly increased global efficiency under both effective butylphthalide. Among metrics, an degree centrality left paracentral lobule related poorer improvement. The model based baseline metrics exhibited high accuracy (88.93%) predicting butylphthalide's efficacy. Conclusion Butylphthalide may restore abnormal organization AD, could be markers therapeutic Clinical Trial Registration This study registered Chinese Registry (Registration Number: ChiCTR1800018362, Date: 2018‐09‐13).

Ischemic stroke (IS) poses a significant threat to global human health and life. In recent decades, we have witnessed unprecedented progresses against IS, including thrombolysis, thrombectomy, few medicines that can assist in reopening the blocked brain vessels or serve as standalone treatments for patients who are not eligible thrombolysis/thrombectomy therapies. However, narrow time windows of thrombolysis/thrombectomy, coupled with risk hemorrhagic transformation, well lack highly effective safe medications, continue present big challenges acute treatment long-term recovery IS. past 3 years, several excellent articles reviewed pathophysiology IS therapeutic based on pathophysiology. Regretfully, there is no comprehensive overview summarize all categories anti-IS drugs/agents designed synthesized molecular mechanisms From medicinal chemistry view point, this article reviews multitude drugs/agents, small molecule compounds, natural products, peptides, others, which been developed mechanism pathophysiology, such excitotoxicity, oxidative/nitrosative stresses, cell death pathways, neuroinflammation, so forth. addition, emerging strategies, nanomedicines, stem therapy noncoding RNAs, recently appeared shortly introduced. Vascular cognitive impairment (VCI) encompasses a range of deficits arising from vascular pathology. The pathophysiological mechanisms underlying VCI remain incompletely understood; however, chronic cerebral hypoperfusion (CCH) is widely acknowledged as principal pathological contributor. Mitochondria, crucial for cellular energy production and intracellular signaling, can lead to numerous neurological impairments when dysfunctional. Recent evidence indicates that mitochondrial dysfunction—marked by oxidative stress, disturbed calcium homeostasis, compromised mitophagy, anomalies in dynamics—plays pivotal role pathogenesis. This review offers detailed examination the latest insights into dysfunction within context, focusing on both origins consequences health. It aims lay robust scientific groundwork guiding development refinement mitochondrial-targeted interventions VCI.

Doxorubicin (DOX) is a widely used chemotherapeutic drug known to cause dose-dependent myocardial toxicity, which limits its clinical potential. DL-3-n-butylphthalide (NBP), substance extracted from celery seed species, has number of pharmacological properties, such as antioxidant, anti-inflammatory, and anti-apoptotic actions. However, whether NBP can protect against DOX-induced acute toxicity still unclear. Therefore, this study was designed investigate the potential protective effects injury underlying mechanism. By injecting 15 mg/kg DOX intraperitoneally, eight-week-old male C57BL6 mice suffered an injury. The treatment group received 80 by gavage once daily for 14 days. To mimic cardiotoxicity DOX, 1uM administered H9C2 cells in vitro. In comparison group, results showed that improved cardiac function decreased serum levels cTnI, LDH, CK-MB. Additionally, HE staining demonstrated attenuated fibrillar lysis breakage DOX-treated mouse hearts. Western blotting assay immunofluorescence suggested oxidative stress, apoptosis, inflammation both vivo Mechanistically, significantly upregulated Nrf2/HO-1 signaling pathway, while Nrf2 inhibitor ML385 prevented protecting myocardium conclusion, Our indicate alleviates activating pathway.
