Accumulating evidence has emerged revealing that noncoding RNAs (ncRNAs) play essential roles in the occurrence and development of hepatocellular carcinoma (HCC). However, complicated regulatory interactions among various ncRNAs HCC are not entirely understood. The newly discovered mechanism competing endogenous (ceRNAs) uncovered different varieties RNAs. In recent years, a growing number studies have suggested ncRNAs, including long circular pseudogenes, major biological functions ceRNA network HCC. These can share microRNA response elements to affect affinity with target RNAs, thus regulating gene expression at transcriptional level both physiological pathological processes. function as ceRNAs involved diverse processes cells, such tumor cell proliferation, epithelial-mesenchymal transition, invasion, metastasis chemoresistance. Based on these findings, act may be promising candidates for clinical diagnosis treatments. this review, we discuss mechanisms research methods networks. We also reviewed advances studying highlight possible directions possibilities diagnostic biomarkers or therapeutic targets. Finally, limitations, gaps knowledge opportunities future discussed.

Hepatocellular carcinoma (HCC) is worldwide accepted most common malignancies, as well the second major cause of death among Chinese with cancer. There an increasing evidence that could prove potential effect long non-coding RNAs (lncRNAs) to biological performance HCC. In present study, high expression level in The Cancer Genome Atlas (TCGA) HCC samples, lncRNA MFI2 Antisense RNA 1 (MFI2-AS1) was closely related poor prognosis and advanced stage patients addition, up-regulation MFI2-AS1 further comfirmed tissues cell line. Ectopic stimulated proliferation metastasis cells, but knockdown suppressed metastasis, indicating exerted oncogenic functions tumorigenesis Simultaneously, compared negative control group, xenograft tumors group were characterized growth, smaller volumes less liver metastases. post-transcriptional regulation FOXM1 by occured mechanistically, playing a role competing endogenous (ceRNA) sponge miR-134. Over-expression increased both at mRNA protein level, whereas it reducd Meanwhile, miR-134 abolished repression shMFI2-AS1 on expression. Furthermore, we demonstrated reverses impact through FOXM1. Collectively, determined crucially acted progression via functioning upregulating expression, conducive promotion better understanding direct diagnostics iatreusiology

Colon cancer is one of the most prevalent malignancies which lead to high occurrence cancer-related deaths. Currently, chemo- and radio- therapies remain primary treatment for advanced colon cancer. Despite initial effectiveness, a fraction patients develops cisplatin resistance, resulting in therapeutic failure. The long non-coding RNA DANCR has been shown be upregulated multiple cancers, indicating an oncogenic role DANCR. This study aims elucidate roles regulating (CDDP) resistance We found was significantly tissues cells compared with normal cells. resistant Moreover, overexpression de-sensitized cisplatin. On other way, silencing dramatically overrode CDDP Bioinformatics prediction revealed could bind seeding region miR-125b-5p as competitive endogenous (ceRNA). interference further validated by luciferase assay. we detected negative correlation between patient tissues: miR-125b clearly downregulated Overexpression sensitized Interestingly, observed were associated increased glycolysis rate. identified enzyme, Hexokinase 2 (HK2) direct target Rescue experiments showed suppressed cellular rate sensitivity through targeting 3'UTR HK2. Importantly, induced miR-125b/HK2 axis, suppression increasement cell. Expectedly, these processes rescued inhibiting DANCR-silenced Finally, demonstrated DANCR-promoted via axis from vivo xenograft mice model. In summary, our reveals new mechanism presenting lncRNA-DANCR-miR-125b/HK2 potential treating chemoresistant

The leading cause of death in pancreatic cancer (PC) patients is the progression metastasis. Recently, long non-coding RNAs (lncRNAs) have been shown to play an important role regulating cell proliferation and metastasis; however, its molecular basis PC remains be explored. In this study, we observed that LINC01094 was markedly overexpressed tissues associated with poor patient prognosis. Downregulation decreased metastasis cells inhibited tumorigenesis mouse xenografts. Mechanically, acted as endogenous miR-577 sponge increase expression target gene, RNA-binding protein lin-28 homolog B (LIN28B), by decoying miR-577, thereby activating PI3K/AKT pathway. Our findings suggest plays critical roles PC, implying can regarded a new biomarker or therapeutic for treatment PC.

Hepatocellular carcinoma (HCC) is a common liver malignancy with high morbidity and poor prognosis. Long non-coding RNAs (lncRNAs) are involved in crucial biological processes of tumorigenesis progression, play four major regulatory roles, namely signal, decoy, guide, scaffold, to regulate gene expression. Through these processes, lncRNAs can target microRNAs (miRNAs) form lncRNA miRNA networks, which cancer cell proliferation, metastasis, drug resistance, the tumor microenvironment. Here, we summarize multifaceted functions networks pathogenesis HCC, potential use diagnostic or prognostic biomarkers, novel therapeutic targets HCC. Metastasis is the primary reason of death in patients with cancer. Small nucleolar noncoding RNAs (snoRNAs) are conserved 60–300 nucleotide RNAs, involved post‐transcriptional regulation mRNAs and RNAs. Despite their essential roles cancer, snoRNAs epithelial‐mesenchymal transition (EMT)‐induced metastasis have not been studied extensively. Here, we used small RNA sequencing to screen for related EMT breast cancer metastasis. We found a higher expression SNORA71A metastatic tissues compared nonmetastatic samples. Additionally, promoted proliferation, migration, invasion MCF‐7 MDA‐MB‐231 cells. Mechanistically, elevated mRNA protein levels ROCK2 , negative regulator TGF‐β signaling. Rescue assays showed abrogated ‐mediated increase EMT. Binding stability regulatory G3BP1, increased half‐life. Furthermore, G3BP1 depletion abolished upregulation . In vivo overexpression tumor growth, knockdown inhibited growth suggest enhances by binding stabilizing
