Duodenal mucosal ablation (DMA) using irreversible electroporation (IRE) with a glucagon-like peptide-1 receptor agonist has been clinically shown to reduce liver lipid deposition in non-alcoholic fatty disease (NAFLD). However, the specific metabolic contributions of DMA IRE NAFLD remain unclear. To assess feasibility and effectiveness rat models. This study aimed to clarify the sequence characteristics and spatiotemporal expression patterns of insulin-like growth factor 1 receptor (IGF1R) gene in Alopex lagopus (Arctic fox), thereby addressing existing knowledge gap regarding IGF1R-mediated regulation this species. The findings establish a crucial foundation for subsequent investigations into correlation between Arctic fox traits. Specific primers were designed based on cDNA canine IGF1R (Accession No. XM_545828). full-length coding (CDS) (1617 bp, encoding 538 amino acids) was successfully cloned via RT-PCR. Phylogenetic analysis using Unweighted Pair Group Method with Arithmetic Mean (UPGMA) algorithm revealed 99% homology canine, confirmed their closest evolutionary relationship. Protein characterization showed that protein has molecular weight 60.62 kDa (theoretical isoelectric point pI = 5.15), containing one fibronectin type-III domain tyrosine kinase domain, classifying it as an acidic hydrophilic transmembrane protein. Phosphorylation site prediction identified 27 phosphorylation sites, secondary structures dominated by α-helices (26.39%) random coils (52.79%). displayed significant tissue-specific variations across 12 examined tissues foxes: highest levels testis, minimal stomach, no detectable duodenum. Spatiotemporal 2-, 4-, 6-month-old individuals, hepatic exhibited progressive increase, testicular reached peak at 6 months, skeletal muscle demonstrated transient upregulation peaking 4 months. Type 2 diabetes (T2D) and obesity are chronic metabolic diseases with global morbidity mortality. Decades of evidence from surgical endoscopic procedures bypassing the duodenum underscore duodenum’s critical role in regulating glycemia body weight. Although surgeries effective, their invasiveness, cost, scalability limit patient access. We developed an orally administered mucin complexing polymeric (MCP) drug, designed to replicate duodenal exclusion physiology. MCPs, intended have electrostatic covalent cross-linkages glycoproteins, form extended network structures resulting alteration mucus barrier properties. Selective targeting is achieved via pH-based activation chemistry. Following screening for physiochemical properties, pharmacokinetics, efficacy, GLY-200 emerged as lead drug candidate replicating physiology improved glycemia, reduced weight, modulation gut hormones rodent models. This targeted oral therapy holds promise treatment T2D by mimicking without invasiveness surgery or procedures.
