During pregnancy the immune system needs to maintain tolerance of foetus while also responding infection, which can cause premature activation inflammatory pathways leading onset labour and preterm birth. The vaginal microbiome is an important modifier birth risk, with Lactobacillus dominance during associated term delivery high microbial diversity increased risk Glycans on glycoproteins along lower female reproductive tract are fundamental microbiota-host interactions mediation responses. However, specific glycan epitopes involved in these processes not well understood. To address this, we conducted glycomic analyses cervicovaginal fluid (CVF) from 36 pregnant women at 4 non-pregnant women. Our analysis N- O-glycans revealed a rich CVF glycome. While were shown be main carriers ABO blood group epitopes, features N-glycans presence abundant paucimannose mannose glycans, remarkable complex bi-, tri-, tetra-antennary glycans decorated fucose sialic acid. We identified immuno-regulatory such as Lewis antigens, found that fucosylation was negatively correlated pro-inflammatory factors, IL-1β, MMP-8, C3a C5a, only sialylated antennae mainly positively those. Similarly, showed positive correlation factors. abundance have previously been hallmarks cancer viral glycosylation, Man8 Man9 glycans. Although each woman had unique profile, longitudinal studies glycosylation consistent throughout who delivered term, whereas experienced extreme exhibited sharp changes glycome shortly before delivery. These findings shed light underlying role maintaining healthy host In addition, discoveries facilitate our understanding has broad implications for women's health.

ABSTRACT Immunometabolism is an emerging growing field that focuses on the role of cellular metabolism in regulation immune cell function and fate. Thus, proliferation, differentiation, activation, populations are modulated by reprogramming their fueling metabolic pathways. Pregnancy entails a fine maternal−fetal interaction to assist energetic demands fetus where trophoblast cells have central role. Maternal neutrophil functional shaping has been proposed though conditioning during pregnancy not studied yet. Here, we explored effects trophoblast‐derived factors rewiring neutrophils from nonpregnant women its impact functions like reactive oxygen species (ROS) production, extracellular trap (NET) release, migration. In parallel, immunometabolic status isolated pregnant (16−20 weeks) was compared with age‐matched control samples. Trophoblast‐derived induced glucose uptake lipid droplet accumulation without activating ROS production or NET release. Conditioned media also inhibited PMA‐induced NETosis partly impairing neutrophils. turn, had increased basal accumulation, women, accompanied higher release NETs. Interestingly, NETs blocked fatty acid oxidation inhibitor indicating contribution activity pregnancy. Results consistent mechanisms underlying point out profiling. These findings provide novel clues understand homeostasis maintenance raise clinical potential monitoring normal complicated pregnancies.

Periodontitis is a potential risk factor for preterm birth (PTB) in women; however, the causal relationship or exact mechanism remain unknown. This study aimed to compare oral microbiome features of mothers with full-term (FTB) those who had delivery.
