Abstract Periodontitis is a dysbiosis‐driven inflammatory disease affecting the tooth‐supporting tissues, characterized by their progressive resorption, which can ultimately lead to tooth loss. A step‐wise therapeutic approach employed for periodontitis. After an initial behavioral and non‐surgical phase, intra‐bony or furcation defects may be amenable regenerative procedures. This review discusses technologies periodontal regeneration, highlighting current limitations future research areas. The search, performed on MEDLINE database, has identified available biomaterials, including biologicals (autologous platelet concentrates, hydrogels), bone grafts (pure putty), membranes. Biologicals have been critically analyzed in terms of composition, mechanism action, clinical applications. Although certain degree regeneration predictable class II defects, complete defect closure hardly achieved. Moreover, treating III remains challenging. key properties required functional are discussed, none commercially biomaterials possess all ideal characteristics. Therefore, needed promote advancement more effective targeted therapies Lastly, improving design reporting studies suggested strictly adhering Consolidated Standards Reporting Trials (CONSORT) 2010 statement.

Background and Objectives: The aim of the present systematic review meta-analysis was to investigate efficacy leukocyte–platelet-rich fibrin (L-PRF) in addition coronally advanced flap (CAF) for treatment both single multiple gingival recessions (GRs) compared CAF alone adjunct connective tissue graft (CTG). Root coverage outcomes using platelet concentrates have gained increased interest. In particular, it has been suggested that adding L-PRF may provide further benefits GRs. Materials Methods: An electronic manual literature search conducted identify randomized controlled trials (RTCs) investigating root with + L-PRF. interest included mean (mRC), recession reduction, keratinized width (KTW) gain, thickness (GT) patient-reported outcome measures (PROms) such as pain perception discomfort. Results: A total 275 patients 611 surgical sites were analyzed. seems show statistically significant results regarding clinical attachment level (CAL) a weighted means (WM) 0.43 95% CI (−0.04, 0.91), p < 0.0001, GT (WM 0.17 (−0.02, 0.36), mRC 13.95 (−1.99, 29.88) alone. Interesting obtained from PRF respect an increase WM 0.07 (−30.22, 30.35), = PPD change 0.26 (−0.06, 0.58), 00001. On other hand, no data when added or combined CTG according (p 0.03 overall). Conclusions: is valid alternative showed discomfort PROms 0.0001). However, remains gold standard treating recession.

To evaluate the efficacy of recombinant human platelet-derived growth factor (rhPDGF)-BB combined with a cross-linked collagen matrix (CCM) for treatment multiple adjacent gingival recession type 1 defects (MAGRs) in combination coronally advanced flap (CAF).Thirty patients were enrolled this triple-blind, randomized, placebo-controlled trial and treated either CAF + CCM rhPDGF, or saline. The primary outcome was mean root coverage (mRC) at 6 months. Complete coverage, gain thickness (GT), keratinized tissue width, volumetric ultrasonographic changes, patient-reported measures also assessed. Mixed-modelling regression analyses used statistical comparisons. At months, mRC rhPDGF alone groups 88.25% 77.72%, respectively (p = .02). A significant GT consistently observed both arms, more so receiving containing through time (0.51 vs. 0.80 mm, on average, p .01). presented greater volume gain, higher soft thickness, superior aesthetic score.rhPDGF enhances clinical, volumetric, outcomes MAGRs above results achieved (ClinicalTrials.gov NCT04462237).

Abstract Objective To evaluate the efficacy of partial-thickness non-advanced tunnel technique (TUN) versus coronally advanced flap (CAF), both combined with a connective tissue graft, in treatment multiple gingival recessions. Materials and methods Twenty-nine patients (83 teeth) affected by recessions were treated two clinical centers either test or control (CAF) intervention graft. Outcomes at 3 6 months after surgery included complete root coverage (CRC—primary outcome), mean (mRC), changes recession depth (RD), probing pocket (PPD), keratinized height (KT). Root sensitivity esthetic score (RES) also evaluated 6-month examination. Surgery duration, wound healing index (WHI), patient-reported outcome measures (PROMs) additionally considered. Results At months, CRC was observed 80.9% 79.5% teeth TUN CAF, respectively (odds ratio = 1.2; p 0.802). No differences between groups terms mRC (TUN 94.0%; CAF 91.1%), RD PPD reductions, sensitivity, RES, WHI. KT increase significantly higher (Difference Means – MD − 1.0 mm; 0.001). duration shorter (MD 19.3 min; 0.001), reported less intra-surgical pain 16.4; 0.028) as well postoperative morbidity compared CAF. Conclusions Both surgical interventions showed similar coverage, albeit associated milder patient’s experience. Clinical relevance techniques have shown for exposed surfaces, although clinicians may consider invasive. Trial registration Clinicaltrials.gov (NCT05122468)
