Abstract
Background
The
molecular
mechanisms
of
osteosarcoma
(OS)
are
complex.
In
this
study,
we
focused
on
the
functions
melanoma
cell
adhesion
molecule
(MCAM),
methyltransferase
3
(METTL3)
and
insulin
like
growth
factor
2
mRNA
binding
protein
1
(IGF2BP1)
in
OS
development.
Methods
qRT-PCR
assay
western
blot
were
performed
to
determine
expression
MCAM,
METTL3,
IGF2BP1
YY1.
MTT

formation
conducted

proliferation.

Abstract
Background
Overconsumption
of
retinoic
acid
(RA)
or
its
analogues/derivatives
has
been
linked
to
severe
craniomaxillofacial
malformations,
such
as
cleft
palate
and
midface
hypoplasia.
It
noted
that
RA
disturbed
the
proliferation
migration
embryonic
palatal
mesenchymal
(EPM)
cells
in
these
yet
exact
mechanisms
underlying
disruptions
remained
unclear.
Methods
A
model
(RA)-induced
fetal
mice
was
successfully
established.
Histological
alterations
were
evaluated
using
Hematoxylin
Eosin
(H&E)
staining
RNA
situ
hybridization
(RNAscope).
Cellular
levels
quantified
via
Cell
Counting
Kit-8
(CCK-8)
assay
EdU
incorporation
assay,
while
cell
capabilities
investigated

Merkel
cell
polyomavirus
(MCPyV)
is
the
foremost
causative
factor
of
carcinoma
(MCC),
a
rare
yet
highly
aggressive
skin
cancer.
Although
evaluation
circulating
IgG
antibodies
against
LT/sT
oncoproteins
clinically
useful
for
MCC
diagnosis/prognosis,
limited
number
assays
identifying
such
have
been
developed.
Herein,
novel
indirect
immunoassay
with
synthetic
epitopes/mimotopes
MCPyV
was
computationally
designed
and
experimentally
validated
on
control
sera
from
healthy
individuals
patients.
Upon
computational
design
five
peptides,
performance
in
detecting
anti-oncoprotein
IgGs
MCPyV-positive
-negative
evaluated.
The
afterwards
extended
individuals,
and,
longitudinal
analysis,
Performance
properties
as
sensitivity
specificity
positive/negative
predictive
values
were
adequate.
Receiver-operating
characteristic
(ROC)
curves
indicated
that
areas
under
(AUCs)
within
low/moderately
accurate
ranges.
