Alzheimer's disease (AD) is a neurological disease, and its signs symptoms appear slowly over time. Although current treatments can alleviate symptoms, they cannot prevent the from progressing. To accurately diagnose treat it therefore necessary to establish effective methods for diagnosis. Apolipoprotein E4 (ApoE4), most frequent genetic risk factor AD, expressed in more than half of patients with making an attractive target AD therapy. We used molecular docking simulations, classical mechanics optimizations, ab initio fragment orbital (FMO) calculations investigate specific interactions between ApoE4 naturally occurring compounds found plant Moringa Oleifera. According FMO calculations, quercetin had highest binding affinity among sixteen because hydroxyl groups generated strong hydrogen bonds residues Trp11, Asp12, Arg15, Asp130. As result, we proposed various derivatives by introducing group into studied their properties. The data clearly showed that adding improved capacity ApoE4. Furthermore, Asp130 were discovered be required significant derivatives. The World Health Organization (WHO) reported that there are 37 million individuals living with the human immunodeficiency virus (HIV) worldwide, majority in South Africa. This chronic disease is managed by effective use of antiretroviral (ARV) drugs. However, prolonged use, ARV drug-induced toxicity remains a clinically complex problem. study investigated drugs on mitochondria and NRF2 antioxidant pathway its possible amelioration using Moringa oleifera Lam (MO) leaf extracts. medicinal plant has range functional bioactive compounds. Liver (HepG2) cells were treated individual drugs: Tenofovir disoproxil fumarate (TDF), Emtricitabine (FTC), Lamivudine (3TC) for 96 h, followed MO extracts 24 h. Intracellular ROS, cytotoxicity, lipid peroxidation, total reduced glutathione (GSH), ATP, mitochondrial polarisation determined. Finally, protein (pNRF2, NRF2, SOD2, CAT, Sirt3) mRNA (NRF2, NQO1 Sirt3, PGC1α) expression measured Western blot qPCR, respectively. TDF, FTC, 3TC significantly increased intracellular ROS extracellular levels both MDA LDH. ARVs also GSH ATP altered polarization. Further, NQO1, UCP2 PGC1α consequently pNRF2, Sirt3 CAT protein. In contrast, was significant reduction LDH post-MO treatment. while increasing GSH, membrane addition to ARV-treated upregulated UCP2, PGC1α, proteins. Extracts from powders of geminated brown rice (Oryza sativa L.), Moringa oleifera, and Cordyceps militaris were combined in various ratios their health-beneficial properties investigated. All extracts exerted the synergistic effects on antioxidant activity as well inhibitions against α-amylase α-glucosidase. Extract formulas T4 (70% germinated jasmine rice, 20% M. oleifera dry leaves, 10% C. militaris) T2 (80% elicited significantly highest inhibition α-glucosidase, respectively. Both exhibited α-amylase. As compared to each other, extract possessed higher glucose consumption-promoting antiinflammatory activities, not toxic normal RAW-264.7 SW-480 human colon cancer cells. Bioactive compounds with health-supporting identified using UHPLC-QTOF-MS/MS. These can be promising therapeutic food.

Abstract Background Aluminum, a well-recognized neurotoxin, is implicated in various neurodegenerative disorders. Moringa oleifera (M. oleifera), known as miracle tree, utilized functional food and nutritional supplement. This study investigates the potential preventive effects of M. extract on aluminum chloride (AlCl3)-induced cortical neurodegeneration rats. Materials methods Therefore, 24 adult male Wistar rats were randomly divided into four distinct groups: negative control, (MOE), AlCl3, AlCl3 + MOE. Treatments administered orally for 28 consecutive days. Cognitive performance, brain oxidative/nitrosative stress, neuroinflammation, apoptotic-cell death, associated histopathological alterations assessed. Results Our results showed that MOE improved spatial learning memory, enhanced antioxidant superoxide dismutase enzyme activity, antagonized nitrosative reduced inflammatory cytokines (tumor necrosis factor-alpha interleukin-6), decreased caspase-3, increased Bcl-2, facilitated repair hippocampal structures. Conclusions We concluded exhibits protective against neurodegeneration, making it promising supplement to counteract aluminum-induced neurotoxic effects.

Aim: The aim of this research is to investigate the potential glucomoringin, derived from Moringa oleifera, as a therapeutic agent for Alzheimer's disease through in silico analysis. Materials & methods: This study employs or computational methodologies, including pkCSM, Swiss ADME, OSIRIS® property explorer, PASS online web resource and MOLINSPIRATION® software, predict pharmacokinetic characteristics biological activity glucomoringin. Results conclusion: Molecular docking indicates strong binding I-1β profile shows cytochrome P450 enzyme inhibition, prompting further dosing strategies.
