Abstract Dietary supplementation with red yeast (RY) has been proposed to enhance immunity and disease resistance in fish; however, its effects tilapia remain underexplored. This study evaluated the impact of Rhodotorula paludigena CM33 on growth performance, immune responses, liver intestinal histology, gene expression, Aeromonas hydrophila tilapia. A total 300 fish were assigned five dietary groups: RY-0 (control), RY-5 (5 g/kg), RY-10 (10 RY-20 (20 RY-40 (40 g/kg). Growth performance was assessed at weeks 4 8. At week 8, blood serum, liver, samples collected for histological expression analyses. Fish then challenged A. an LD₅₀ dose 10⁵ CFU/fish via intraperitoneal injection. Results showed that R. significantly improved polynomial regression identifying ~ 24 g/kg as optimal, experimental fed 20 achieved highest weight, gain, feed efficiency. Antioxidant responses enhanced, evidenced by increased ABTS radical scavenging activity, superoxide dismutase upregulation immune-related antioxidant genes (IL1, MHC, LBP, GSR, GPX, IGF, HSP70, TNF). Histological analysis revealed enhanced structure hepatocyte integrity, exhibiting elongated villi reduced hepatic vacuolation compared control. Bacillus sp. THPS1 is a novel strain isolated from high-temperature hot spring in Thailand, exhibiting distinctive genomic features that enable adaptation to an extreme environment.
