Prevotella copri is an abundant member of the human gastrointestinal microbiome, whose relative abundance has curiously been associated with positive and negative impacts on diseases, such as Parkinson’s disease rheumatoid arthritis. Yet, verdict still out definitive role P. in health, effect different diets its gut microbiome. The puzzling discrepancies among studies have only recently attributed to diversity strains, which substantially differ their encoded metabolic patterns from commonly used reference strain. However, strain differences cannot be resolved by common 16S rRNA amplicon profiling methods. Here, we scrutinize copri, versatile potential, hypotheses behind conflicting observations association diet health. We also provide suggestions for designing bioinformatics pipelines better research copri.

Gut microbiota regulates the immune system, development and progression of autoimmune diseases (AIDs) overall health. Recent studies have played a crucial part in understanding specific role different gut bacterial strains their metabolites AIDs. Microbial signatures AIDs are revealed by advanced sequencing metabolomics studies. Microbes such as Faecalibacterium prausnitzii, Akkermansia muciniphila, Anaerostipes caccae, Bacteroides sp. , Roseburia Blautia faecis Clostridium lavalense Christensenellaceae Coprococcus Firmicutes Ruminococcaceae Lachnospiraceae Megamonas sp., Monoglobus Streptococcus pneumoniae Bifidobacterium help maintain homeostasis; whereas, Prevotella copri, Ruminococcus gnavus, Lactobacillus salivarius, Enterococcus gallinarum, Elizabeth menigoseptica, Collinsella Escherichia Fusobacterium Enterobacter ludwigii, Enterobacteriaceae Proteobacteria Porphyromonas gingivalis nigrescens Dorea cause immuno-pathogenesis. Abstract Background Gut microbiota has been extensively demonstrated to modulate host lipid metabolism. Higher intramuscular fat (IMF) accumulation in Chinese indigenous breed pigs is associated with their special gut structure. However, the specific microbes and metabolic pathways responsible for deposition are still poorly understood. Results In present study, a comparative analysis of metabolome obese Ningxiang (NX) lean Duroc × Landrace Yorkshire (DLY) was conducted. The results revealed higher abundance lactobacilli correlation branched-chain amino acid (BCAA) metabolism pathway NX pigs. We proceeded verify roles various strains originating from BCAA lipids SD rats. that L. reuteri fundamental species modulating increased circulating levels positively linked greater deposition. Additionally, it verified ability improve synthesis enhance IMF content DLY expression genes related also significantly upregulated. Conclusions Taken together, our imply pig-derived regulates plays potential role improving meat quality pig breeds through modulation provide new strategy commercial by influencing supplementing dietary additives.

This was the first study that examined effects of oat β-glucan and inulin on diet-induced nonalcoholic steatohepatitis (NASH) in circadian-disrupted (CD)-male C57BL/6J mice. CD intensified NASH, significantly increasing alanine aminotransferase upregulating hepatic tumor necrosis factor α (TNFα) transforming growth β 1 (TGFβ1). However, these observations were alleviated by treatments. Compared to NASH mice, decreased liver index, aspartate (AST), insulin. In prebiotic-treated significant negative correlations found between enrichment Muribaculaceae bacterium Isolate-036 (Harlan), Isolate-001 (NCI), Bacteroides ovatus after supplementation with TNFα TGFβ1 levels; Isolate-110 (HZI) AST level. conclusion, exhibited similar antiliver injury, anti-inflammatory, antifibrotic activities but had no effect cecal short-chain fatty acids gut microbiota diversity

Abstract Non‐alcoholic fatty liver disease (NAFLD) represents a growing global health concern that can lead to and cancer. It is characterized by an excessive accumulation of fat in the liver, unrelated alcohol consumption. Studies indicate gut microbiota‐host crosstalk may play causal role NAFLD pathogenesis, with epigenetic modification serving as key mechanism for regulating this interaction. In review, we explore how interplay between microbiota host epigenome impacts development NAFLD. Specifically, discuss microbiota‐derived factors, such lipopolysaccharides (LPS) short‐chain acids (SCFAs), modulate DNA methylation histone acetylation genes associated NAFLD, subsequently affecting lipid metabolism immune homeostasis. Although current literature suggests link development, our understanding molecular mechanisms signaling pathways underlying remains limited. Therefore, more comprehensive epigenomic multi‐omic studies, including broader clinical animal experiments, are needed further linking NAFLD‐associated genes. These studies anticipated improve microbial markers based on strategies provide novel insights into pathogenesis ultimately addressing significant unmet need.

Increased abundance of Segatella copri (S. copri) within the gut microbiota is associated with systemic inflammatory diseases, including rheumatoid arthritis. Although outer-membrane vesicles (OMVs) Gram-negative bacteria are important players in microbiota-host communication, effect S. copri-derived OMVs on immune cells unknown. Macrophages engulf and eliminate foreign material conditioned by environmental signals to promote either homeostasis or inflammation. Thus, we aimed explore impact copri-OMVs human macrophages vitro, employing THP-1 monocyte-derived macrophage models. The uptake DiO-labeled into was monitored confocal microscopy flow cytometry. Furthermore, phenotype cytokine secretion naïve (M0), pro-inflammatory (M1), anti-inflammatory (M2) analyzed cytometry ELISA, respectively. We show that enter through macropinocytosis clathrin-dependent mechanisms. copri-OMVs, but not parental bacterium, induced a dose-dependent increase expression M1-related surface markers M0 M2 activated large amounts cytokines M1 macrophages. These results highlight an role promoting responses, which might contribute diseases.
