Background/Objectives: Transient Receptor Potential Melastatin 8 (TRPM8) is a non-selective, Ca2+-permeable cation channel involved in thermoregulation and other physiological processes, such as basal tear secretion, cell differentiation, insulin homeostasis. The activation deactivation of TRPM8 occur through genetic modifications, interactions, signaling cascades. Recent evidence suggests significant role the hypothalamus amygdala related to pain sensation sexual behavior. Notably, has been implicated neuropathic pain, migraines, neurodegenerative diseases Parkinson’s disease. Our laboratory identified testosterone high-affinity ligand TRPM8. deficiency appears influence behavioral traits mice, like increased aggression deficits satiety. Here, we aim explore pathways altered brain tissues TRPM8-deficient mice using expression methylation profiles messenger RNA (mRNA) long non-coding (lncRNA). Specifically, focused on regions integral hormonal control, including olfactory bulb, hypothalamus, amygdala, insula. Methods: was isolated purified for microarray analysis collected from male wild-type knockout mice. Results: We various differentially expressed genes tied multiple pathways. Among them, androgen–estrogen receptor (AR-ER) pathway, steroidogenesis reward cocaine pathway are particularly worth noting. Conclusions: These results should bridge existing gaps knowledge regarding inform potential targets future studies elucidate its behavior changes pathology associated with activity.

The direct pathway from the lateral hypothalamus to mouse olfactory bulb (OB) includes neurons that express neuropeptide orexin-A, and others do not. OB-projecting not orexin-A are present in an area of known contain melanin-concentrating hormone (MCH). We used virally mediated anterograde tract tracing immunohistochemistry for MCH demonstrate OB is broadly innervated by axon projections both populations neurons. Orexin-A were expressed each layer across its anterior posterior axis. Both genetically heterogeneous, with subsets co-express isoform vesicular glutamate transporter (VGLUT). high-resolution confocal imaging test whether reflect this glutamatergic heterogeneity. majority (∼57%) putative terminals overlapped VGLUT2, smaller proportions co-expressed VGLUT1, or did overlap either VGLUT1 VGLUT2. In contrast, only ∼26% overlapping VGLUT. Therefore, heterogeneous include can release two different neuropeptides. themselves distinct ratios non-glutamatergic terminals. A projection orexin-A-expressing was confirmed using tracing.Orexin-A (MCH), neuropeptides primarily produced throughout OB.Approximately 64% type 1 (VGLUT1) 2 (VGLUT2). 25%

Although the metabolic state of an organism affects olfactory function, precise mechanisms and their impact on behavior metabolism remain unknown. Here, we assess whether ghrelin receptors (GHSRs) in bulb (OB) increase function influence foraging behaviors metabolism. We performed a detailed behavioural analysis mice lacking GHSRs OB (OBGHSR deletion). also analsyed scRNA-seq spatial transcriptomic datasets to GHSR+ cells main accessory bulbs, as well anterior nucleus. OBGHSR deletion affected discrimination habituation both food non-food odors. Anxiety-like depression-like were significantly greater after deletion, whereas exploratory was reduced, with greatest effect under fasted conditions. impacted feeding evidenced by altered bout number duration, buried food-seeking. increased body weight fat mass, spared utilisation chow diet impaired glucose indicating dysfunction. Cross referenced revealed glutamate neurons Ablation reduced ghrelin-induced finding phenocopied results seen deletion. OBGHSRs help maintain particularly during hunger, facilitate behavioral adaptations that optimise food-seeking anxiogenic environments, priming pathways preparation for consumption.

Adequate intake of dietary essential amino acids (eAAs) is vital for protein synthesis and metabolism. Any single eAA deprivation sufficient to increase in Drosophila melanogaster. Vasopressin (VP) plays a crucial role in social memory even at the level of olfactory bulb (OB), where OB VP cells are activated during interactions. However, it remains unclear how modulates processing to enable enhanced discrimination very similar odors, e.g., rat body odors. Thus far, has been shown that reduces firing rates mitral (MCs) odor presentation vivo and decreases amplitudes nerve-evoked excitatory postsynaptic potentials (ON-evoked EPSPs) external tufted vitro . We performed whole-cell patch-clamp recordings population Ca 2+ imaging on acute slices. recorded ON-evoked EPSPs as well spontaneous inhibitory currents (IPSCs) from two types projection neurons: middle (mTCs) MCs. bath application reduced frequencies IPSCs mTCs but did not change those Therefore, we analyzed interneurons, i.e., periglomerular (PGCs) granule (GCs), search for origin increased inhibition mTCs. increase evoked either type interneurons. next two-photon glomerular layer superficial GC responses stronger ON stimulation than experiments should evoke action measured cells. observed influx juxtaglomerular somata. Thus, our findings indicate by neurons via strong input-mediated interneuron activity. This neural modulation could improve representation hence, better discriminability conspecific
