Background/Objectives: Proliferative verrucous leukoplakia (PVL) is an oral disorder with the greatest degree of malignant transformation. However, it relatively rare. This study compared clinical characteristics patients squamous cell carcinoma (OSCC) who had and not been previously diagnosed PVL. Methods: case control classified as early (T1 T2) or advanced (T3 T4) OSCC according to T size Tumor, Node, Metastasis (TNM) classification, including age, gender, location, type carcinoma. The analysis involved 140 patients. Group 1: 50 PVL (OSCC-PVL) 2: 90 without (OSCC-noPVL). Results: OSCC-PVL were younger than those OSCC-noPVL, but this did reach statistical significance. Regarding patient much more frequently female (70%), while OSCC-noPVL was prevalent in men (65.5%) (p < 0.01). There also significant differences locations between two groups: gingiva most tongue OSCC-noPVL. Erythroleukoplastic forms significantly common (30% vs. 7.7%), ulcerated frequent (63.3% 42%). Finally, stages our OSCC-PVL. Conclusions: We found that preceded by women, less aggressive forms,

Background/Objectives: Oral cancers in patients with proliferative verrucous leukoplakia (PVL-OSCC) exhibit different clinical and prognostic outcomes from those seen conventional oral squamous cell carcinomas (cOSSCs). The aim of the present study is to compare genome-wide DNA methylation signatures fresh frozen tissues between PVL cOSCC using Illumina Infinium MethylationEPIC BeadChip. Methods: This case–control was carried out at Stomatology Maxillofacial Surgery Department General University Hospital Valencia. For epigenomic study, unsupervised exploratory bioinformatic analyses were performed principal component heatmap analysis. Supervised differential conducted a rank-based regression model penalized logistic identify potential biomarkers. Results: global profiles did not allow us differentiate distinct cancer groups. However, two supervised confirmed existence carcinoma phenotypes. We identified 21 differentially methylated CpGs corresponding 14 genes. Among them, three had been previously assigned any known gene, remaining associated genes unrelated cancer. AGL, WRB, ARL15 as Conclusions: emphasizes significant role epigenetic dysregulation OSCC, particularly cases preceded by PVL. have provided data on that could be involved molecular carcinogenesis PVL-OSCC.

Oral potentially malignant disorders (OPMDs) are a group of conditions that carry risk oral squamous cell carcinoma (OSCC) development. Recent studies indicate periodontal disease-associated pathogenic bacteria may play role in the transition from healthy mucosa to dysplasia and OSCC. Yet, microbial signatures associated with have not been established. To characterize at these different sites, we performed 16S sequencing analysis both swab formalin-fixed, paraffin-embedded tissue (FFPE) samples. We collected swabs (from patients), histologically normal adjacent dysplasia, low-grade dysplasia. Additionally, FFPE samples OSCC, plus low grade high-grade were also collected. The data demonstrate significant differences alpha beta diversities sites mucosa, as well increased dissimilarities within sites. found Proteobacteria phyla abundance increased, concurrent progressive decrease Firmicutes abundance, altered levels Enterococcus cecorum, Fusobacterium periodonticum, Prevotella melaninogenica, canifelinum when moving diseased Moreover, sample indicates microbiome be areas normal, including Furthermore, trends specific changes preceded those tissues, signifying early detection opportunities for clinical diagnosis. In addition, evaluated gene expression profile OSCC cells (HSC-3) infected either P. gingivalis, T. denticola, F. nucelatum, or S. sanguinis three periodontopathogens enrich genetic processes related cancer progression, skin keratinization/cornification, while commensal enriched RNA processing adhesion. Finally, reviewed literature bacteria, such Streptococci genus, simultaneous increase mainly Bacteroidetes genus. These findings suggest features can serve novel biomarkers disease progression.

Abstract Objective Proliferative verrucous leukoplakia (PVL) is considered a clinically distinct entity from other oral leucoplakias (OLs) due to its clinical presentation and evolution. However, molecular differences between them remain unclear. We aimed determine whether there are methylation PVL forms of OLs. Materials Methods Oral biopsies 12 patients with PVL, eight homogeneous leucoplakia (HL), 10 healthy individuals were obtained for genome‐wide DNA analysis via the Infinium EPIC Platform. Results A total 1815 differentially methylated CpGs found HL, prominent state hypermethylation in HL patients. covered 813 genes roles, including cell adhesion, extracellular matrix organization, synaptic signaling. 43% these had been previously described cancer associated prognosis. developed multinomial logistic regression model able differentiate control samples.
