Abstract Objectives: Clinical studies have confirmed that galectin-3 (Gal-3) levels are significantly elevated in periodontitis patients. The present study aimed to explore the effects of Gal-3 inhibition on periodontal inflammation vitro and vivo. Methods: Human gingival fibroblasts (HGFs) with or without knockdown were stimulated by lipopolysaccharide (LPS), a ligation-induced mouse model treated inhibitor was established. Hematoxylin–eosin (H&E) immunohistochemistry (IHC) staining used evaluate tissues. Quantitative real-time polymerase chain reaction (qRT-PCR) enzyme-linked immunosorbent assay (ELISA) detect Gal-3, interleukin (IL)-6, IL-8, C-C motif ligand 2 (CCL2) expression. Immunofluorescence western blotting NF-κB ERK signaling pathway activation. Micro-computed tomography analyse degree bone loss. Results: up-regulated inflamed tissues LPS-induced HGFs. markedly decreased IL-6, CCL2 expression blocked activation In model, alleviated IL-1β IL-6 infiltration tissue mitigated Conclusions: notably partly through blocking

Putative associations between certain bacteria and gastric cancer have been described previously; however, the mechanisms underlying such are not thoroughly understood. We aimed to confirm presence of Fusobacterium nucleatum in biopsy samples patients with gastroduodenal diseases, including cancer, concomitant or without Helicobacter pylori infection. Three hundred disorders, gastritis, duodenal ulcer, were included. One eighty (60%) negative 120 (40%) positive for H. pylori. Associations age, gender, other demographics statistically insignificant (p > 0.05). The prevalence F. infection was determined two patient categories: 215 (72%) nucleatum; these cases, 95 showed evidence of, lacked, Gastric biopsies obtained from but revealed a significant

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline and neuronal loss, affecting millions worldwide. Emerging evidence highlights the oral microbiome—a complex ecosystem of bacteria, fungi, viruses, protozoa as significant factor in health. Dysbiosis microbiome contributes to systemic inflammation, disrupts blood–brain barrier, promotes neuroinflammation, processes increasingly implicated pathogenesis AD. This review examines mechanisms linking dysbiosis through oral–brain oral–gut–brain axis. These interconnected pathways enable bidirectional communication between cavity, gut, brain via neural, immune, endocrine signaling. Oral pathogens, such Porphyromonas gingivalis, along with virulence factors, including lipopolysaccharides (LPS) gingipains, contribute while metabolic byproducts, short-chain fatty acids (SCFAs) peptidoglycans, further exacerbate immune activation. Additionally, this explores influence external diet, pH balance, medication use, smoking, alcohol consumption, hygiene, on microbial diversity stability, highlighting their role shaping outcomes. The dynamic interplay gut microbiomes reinforces importance homeostasis preserving neurological interventions, probiotics, prebiotics, dietary modifications, offer promising strategies support function reduce risk diseases, AD, maintaining diverse microbiome. Future longitudinal research needed identify long-term impact cognition.

The gingival mucosal barrier, an important oral cavity plays a significant role in preventing pathogenic microorganism invasion and maintaining periodontal tissue health. Pathogenic of the mucosa produces large number cytokines. Among them, pyroptosis is player exacerbating immune-inflammatory responses, leading to destruction. However, mechanism immune response it triggers have not been fully elucidated. We provide overview recent advances understanding physical barrier inflammation-induced hyperimmunity.
