ABSTRACT Objectives Oral leukoplakia (OL), oral lichen planus (OLP), and submucous fibrosis (OSF) are precancerous lesions known to progress squamous cell carcinoma (OSCC). Understanding the early molecular events driving oral carcinogenesis is vital for diagnosing squamous cell carcinoma (OSCC) promptly. While metabolic differences between leukoplakia (OLK), OSCC, and healthy mucosa have been reported, changes distinguishing malignant transformed OLKs (MT-OLK) from non-transformed (NT-OLK) remain unexplored. Here, we examine metabolomic profiles of 5 cases MT-OLK 15 NT-OLK to identify key predictive molecules using untargeted high-performance liquid chromatography-mass spectrometry. The potentially discriminant compounds were highlighted through a robust statistical analysis workflow, dysregulated pathways illustrated by enrichment analysis. Seventeen features, primarily lipids-including phospholipids, oxidised lipids, cholesteryl esters, fatty acids-were identified as discriminants across bioinformatic approaches. Pathway revealed alterations in lipid metabolism, particularly acid synthesis degradation, steroid hormone biosynthesis, glycerophospholipid metabolism. Predictive models showed high accuracy (AUC = 0.88) two groups. This study suggests that metabolomics has potential differentiate identifying candidate biomarkers may contribute understanding transformation. Validation larger cohorts warranted translate these findings into clinical practice.

Objectives The purpose of this systematic review and meta-analysis was to appraise, both quantitatively qualitatively, the extant evidence regarding role EGFR upregulation in predicting malignant transformation risk associated with oral potentially disorders (OPMD). ABSTRACT Objectives Oral epithelial dysplasia (OED) can be graded histopathologically by using a binary‐ or three‐tiered grading system. This study aimed to analyze and compare the prognostic value of two systems in large single‐institute material. Materials Methods The was retrospective histopathological re‐evaluation analysis 612 OEDs over 29 years at Institute Dentistry, University Turku, Finland. OED patients who later developed squamous cell carcinoma (OSCC) were identified from Finnish Cancer Registry database. Results Altogether, mean age 59.9 years. male:female ratio 0.67. According original diagnosis, 67.7% samples mild dysplasia, 28.7% moderate 3.6% severe cases. With binary‐tiered low‐grade 84.1% more common than high‐grade 15.9%. malignant transformation rate 7.8%. Regarding oral SCC development, no difference found between predicting progression ( p = 0.060). Conclusions Both classification had similar predictive for transformation.
