Numerous studies indicate that periodontal diseases affect a large portion of the adult population, and gum disease is primary cause losing teeth in future. Periodontal are global health concern. most common serious oral illnesses, affecting not only gums but also tissue bone support teeth. It begins as gingivitis, slight swelling resulting from buildup bacterial plaque. If treated appropriately, it may advance to periodontitis, where inflammation infection spread deeper, causing damage bone. Periodontitis, a common dental illness, causes periodontal tissue inflammation and irreversible bone loss, inevitably resulting in tooth loss. Hyperhomocysteinaemia (HHcy), defined as blood total homocysteine (Hcy) levels greater than 15 µmol/L, is linked to increased cardiovascular disease risk. Mounting evidence indicates connection between HHcy periodontitis; however, the underlying processes remain unknown. Herein, we explored mechanisms by which exacerbates osteoclast formation. In an animal model of periodontitis treated with HHcy, attachment loss was aggravated, both systemic gingival tended increase; additionally, antioxidant-related proteins were suppressed expressed at low levels, whereas oxidative damage-related protein expression increased. RAW264.7 cells LPS or + Hcy, Hcy group presented reactive oxygen species (ROS) fluorescence intensity, Nrf2/HO-1 signalling pathway suppression associated inflammatory cytokine (TNF-α) expression. monocyte osteoclasts Rankl suppression, increase osteoclast-related (NFATc-1 CTSK), more pronounced osteoclastic phenotype. ABSTRACT Aim Circulating platelets are essential in hemostasis, thrombosis, and immune responses. Modifications platelet function may impact immunological reactions to dental biofilm cardiovascular health. Understanding changes status activity patients with periodontitis is still a subject of investigation. This study aimed synthesize evidence from observational studies that investigated without periodontitis. Methods MEDLINE‐PubMed, EMBASE, Cochrane‐CENTRAL Library databases were searched up November 2024. Primary outcomes included count (PC) mean volume (MPV). Secondary encompassed any other biomarker relevant activity. Methodological quality was evaluated using the Newcastle‐Ottawa scale, heterogeneity analyzed. Descriptive analysis meta‐analysis, incorporating trial sequential PC MPV, conducted. The body graded by utilizing Grading Recommendations Assessment, Development, Evaluation (GRADE). Background/Objectives: Periodontal disease involves chronic immunoinflammatory processes and microbial dysbiosis, making phytochemicals with anti-inflammatory properties potential therapeutic agents. This study aimed to assess the modulatory effects of yellow passion fruit bagasse extract (PFBE) on periodontal cells under condition. Methods: A human ligament (PDL) cell line was exposed F. nucleatum ATCC 25586 simulate a environment in vitro presence absence PFBE containing three different concentrations (0.25, 0.50, 1.00 µg/mL) piceatannol. Pro-inflammatory markers (TNF-α, IL-8, CCL2), antioxidant enzyme SOD2, protease marker MMP-1 were analyzed by real-time PCR. Protein levels assessed via ELISA NF-κB nuclear translocation immunofluorescence. Cell viability investigated using live/dead alamarBlue assays, wound healing evaluated an automated scratch assay. Results: PDL significantly increased gene protein expression all inflammatory markers. The stimulatory reduced when simultaneously PFBE. triggered while abrogated nucleatum-stimulated at 60 min. Abstract The primary objective is to critically analyze approaches local therapy for periodontal disease, particularly in light of its well‐established systemic implications. We specifically address common misconceptions and overestimations regarding the potential treatments contrast their actual clinical effectiveness. Historically, therapeutic approach periodontitis has concentrated on intervention within pockets. Local drug delivery systems show targeting infections with high concentrations while minimizing side effects. This could potentially help overcome challenges posed by microbial biofilms. However, evidence suggests that not a disease affecting only confined areas dentition. Currently, presence multiple deep pockets, less cost‐effective than antibiotics due costs products extensive professional chair time required application. Isolated pockets otherwise stable dentitions are often caused problems cannot be resolved antimicrobial agents. Finally, recurrent biofilm formation residual during maintenance phase can addressed through suitable physical rather chemical procedures.
