Misuse and overuse of antibiotics in aquaculture has proven to be an unsustainable practice leading increased bacterial resistance. An alternative strategy involves the inclusion immunostimulants fish diets, especially fungal herbal compounds already authorized for human consumption, hence without environmental or public health concerns. In this study, we used a holistic cross-disciplinary pipeline assess immunostimulatory properties two fungi: Trametes versicolor Ganoderma lucidum; one supplement, capsaicin form Espelette pepper (Capsicum annuum), combination these additives on rainbow trout (Oncorhynchus mykiss). We investigated impact diet supplementation 7 weeks survival, growth performance, cellular, humoral, molecular immune parameters, as well intestinal microbial composition fish. Uptake influenced expression related genes, generating inflammatory response. Significant differences were detected spleen-tlr2 gene expression. Supplementation with correlated structural changes microbiota enhanced overall diversity. Results demonstrated that different treatments had no adverse effect performance suggesting safety feed at tested concentrations. While mechanisms multifactorial interactions remain unclear, study provides insights not only regard nutrition compounds, but also how combined gut approach can shed light efficacy immunostimulant potential commercial supplements.

Abstract Fish meal (FM) replacement is essential for the sustainable expansion of aquaculture. This study focussed on feasibility replacing FM with a single‐cell protein (SCP) derived from methanotrophic bacteria ( Methylococcus capsulatus , Bath) in barramundi fry Lates calcarifer ). Three isonitrogenous and isoenergetic diets were formulated 0%, 6.4% 12.9% inclusion SCP, by 25% 50%. Barramundi (initial body weight 2.5 ± 0.1 g) fed experimental 21 days to assess growth performance, gut microbiome composition histopathology. Our findings revealed that both levels SCP induced detrimental effects fry, including impaired reduced survival compared control group (66.7% 71.7% 50%, respectively; p < .05). Both dietary treatments presented mild necrotizing enteritis subepithelial oedema accumulation PAS positive, diastase resistant droplets within hepatocytes (ceroid hepatopathy) pancreatic atrophy. Microbiome analysis marked shift microbial community potential opportunistic genus Aeromonas .
