ABSTRACT Emerging infectious diseases are a major threat to global aquaculture production and projected increase in frequency due anthropogenic stressors climate change. Our ability screen the aquatic ‘infectome’ is dramatically improving through unbiased metatranscriptomic (i.e., total RNA) sequencing. Despite recent advances molecular diagnostics next‐generation sequencing, outbreak investigation often encounters limitations targeted nature of conventional techniques, which may only capture small portion diverse array disease‐causing agents present nature, sometimes requiring years resolve. Abstract Vibrio species are an emerging public and animal health risk in marine environments the opportunistic bacterial pathogen harveyi is a major disease for tropical aquaculture. Current understanding of virulence V. limited by strain-specific variability complex host-pathogen dynamics. This study sought to integrate genomic investigation, phenotypic characterisation vivo challenge trials barramundi ( Lates calcarifer) increase our virulence. We identified two hypervirulent isolates, Vh-14 Vh-15 that caused 100% mortality fish within 48 hours, were phenotypically genotypically distinct from other isolates. Virulent isolates contained multiple plasmids, including 105,412 bp conjugative plasmid with type III secretion system genes originally Yersinia pestis . The emergence this plasmid-mediated patho-variant poses potential threat sustainable production finfish Southeast Asia, Mediterranean Australia. In addition, we observed effect temperature on indicators activity at 28°C 34°C compared 22°C. suggests fluctuations associated climate change may act as stressor bacteria, increasing gene host adaptation. ABSTRACT Lactococcosis, primarily caused by Lactococcus garvieae , is prevalent in various freshwater fish species globally. Yet, its significance Thai aquaculture remains poorly understood. This study investigated Lactococcosis‐associated diseases across four red tilapia ( Oreochromis sp.) and one Asian sea bass Lates calcarifer ) farm Thailand 2022. Chronically diseased displaying clinical signs such as exophthalmia or corneal opacity, erratic swimming, scale loss skin haemorrhage, well apparently healthy specimens, were studied. Bacterial isolation was performed from the internal organs. Gram‐positive bacterial isolates selected later identified L. lactis n = 4), taiwanensis 1), 2) petauri based on 16S rDNA gyrB partial sequences. Sub‐adults of Nile intracoelomic (IC) injected with representatives each three at varying doses (10 3 –10 7 CFU/fish), while juvenile IC ranging 10 2 to 8 CFU/fish. Control groups received culture medium alone. By end 21‐day test, mortality rates all challenge ranged 0% 14.82%. Some observed challenged higher bacteria. Enzyme‐linked immunosorbent assay (ELISA) showed that survivor exposed high elicited specific antibodies IgM. In summary, this identifies presence farmed fishes, which exhibit low virulence under laboratory‐controlled challenges. These appear be opportunistic rather than pathogenic, potentially indicating underlying health challenges within affected farms.

Anthropogenic climate change is altering coastal systems globally, affecting macroalgae abundance and composition. These host diverse microbiomes, including pathogenic bacteria. Of particular concern are Vibrio species, such as parahaemolyticus vulnificus , which linked to human disease impact public health, the economy, recreation in areas. This study examined presence of spp. across seven genera a temperate estuary (Narragansett Bay, RI, USA). Using colony-forming unit (CFU) counts multiplex qPCR, we quantified V. assess if varied by genus morphology. We also potential environmental factors influencing prevalence. demonstrate that both were present on all genera, with showing higher average abundance. Abstract Aquaculture plays a key role in global food security, yet disease outbreaks remain major challenge. While host transcriptomic response have been widely studied, the of microbial community risk aquaculture remains understudied. Here, we conducted metatranscriptomic analysis gill microbiomes Atlantic salmon ( Salmo salar ) by repurposing publicly available RNA-seq data originally generated for transcriptomics. These poly(A)-selected datasets were collected prior to outbreak, allowing us examine functional patterns retrospectively. Taxonomic profiles Bracken and MATAM showed strong concordance across dominant genera, confirming that retain sufficient signal downstream classification. In addition tank-dependent variation, our presence-absence revealed survival-associated microbiome shifts. Several taxa, including Sedimentibacter , Fusarium Demequina more observed non-survived individuals, whereas Micrococcaceae, Pseudomonas, Burkholderiaceae enriched survivors. Functional gene distinct transcriptional responses within same taxa between survivors non-survivors, particularly metabolic adaptation, stress response, host-microbe interactions. results suggest host-derived datasets, though not designed analysis, offer new opportunities investigate early signals environmental factors associated with susceptibility aquaculture.
