SUMMARY RNA‐Sequencing is widely used to investigate changes in gene expression at the transcription level plants. Most plant RNA‐Seq analysis pipelines base normalization approaches on assumption that total transcript levels do not vary between samples. However, this has been demonstrated. In fact, many common experimental treatments and genetic alterations affect efficiency or RNA stability, resulting unequal abundance. The addition of synthetic controls a simple correction for variation mRNA levels. adding spike‐ins appropriately challenging with complex tissue, carefully considering how they are added essential their successful use. We demonstrate external as control produces differences compared traditional methods, even two times day untreated illustrate use 3' present pipeline accounts transcriptional evaluate effect methods identifying differentially expressed genes context time response chilling stress sorghum.

SUMMARY Water availability is a major determinant of crop production, and rising temperatures from climate change are leading to more extreme droughts. To combat the effects on yields, we need develop varieties that tolerant water‐limited conditions. We aimed determine how diverse types (winter/spring oilseed, tuberous, leafy) allopolyploid Brassica napus , species contains economically important rapeseed oilseed crop, respond prolonged water limitation. exposed plants an 80% reduction in assessed growth color high‐throughput phenotyping system over 4 weeks ended experiment with tissue collection for time course transcriptomic study. found overall across cultivars but varying degrees. Diel transcriptome analyses revealed significant accession‐specific changes time‐of‐day regulation photosynthesis, carbohydrate metabolism, sulfur metabolism. Interestingly, there was extensive variation which homoeologs two parental subgenomes responded limitation could be due differences regulatory regions these lines. Follow‐up experiments select confirmed maintained photosynthetic health during while slowing growth. In examined, day levels glucosinolates, sulfur‐ nitrogen ‐rich specialized metabolites, consistent diel responses. These results suggest lines adjusting their stores under conditions through distinct regulation.

Influence of sowing date and cultivar on grain and protein yield of grain sorghum [Sorghum bicolor (L.) Moench] DOI

Опубликована: Ноя. 1, 2024

Abstract Sorghum is a warm-climate crop that tolerant of water shortages and drought. In terms climate change in Europe, this cereal could be increasingly grown to replace maize where cannot grown. Unfortunately, the agronomics especially optimum sowing date are currently not sufficiently researched, which means many farmers potentially interested growing sorghum do know what use. The diversity cultivar also poses problem their appropriate selection for weather conditions Poland. study used two different dates sorghum: first decade May second May. Three grain cultivars were used: Albanus, Anggy GK Emese. conducted experiment, such features as yield its components as: plant density, number panicles, tillering index, weight per panicle thousand mass studied. Protein content was studied protein calculated. Emese variety proved most stable yield, but it sensitive date. other varieties Albanus showed varied response yield.
