Pu’an County, located in southwestern Guizhou Province, China, is one of the original habitats for wild tea plants. It renowned not only as “Home Ancient Tea Trees China” but also “Core Production Area High-Quality Early China”. The ancient Camellia tetracocca trees are considered “living fossil”. Understanding genetic diversity C. Pu’an, Guizhou, great significance addressing conservation concerns and mitigating erosion this endemic species. This study investigates population structure plants County to support development strategies. We genotyped 138 specimens using 40 intron-length polymorphism markers. A total 180 alleles were detected, with allele numbers per locus ranging from 2 10 an average 4.50. number effective varied 1.36 8.01, 2.86. Shannon information index ranged 0.28 2.19, 1.10. Nei’s gene 0.14 0.88, 0.58. polymorphic content (PIC) 0.85, Our findings indicate that high. Specifically, Qingshan Township surpassed Xindian Township. Analysis molecular variance indicated 91.59% variation occurred within subpopulations, suggesting limited differentiation. Despite their geographical separation, populations showed a complex relationship (FST = 0.04). STRUCTURE analysis identified three distinct clusters, indicating demographic history. These underscore highlight need strategies prioritize protection genetically diverse especially region.
