ABSTRACT Aging‐related decreases in cardiac and skeletal muscle function are strongly associated with various comorbidities. Elamipretide (ELAM), a novel mitochondria‐targeted peptide, has demonstrated broad therapeutic efficacy ameliorating disease conditions mitochondrial dysfunction across both clinical pre‐clinical models. Herein, we investigated the impact of 8‐week ELAM treatment on pre‐ post‐measures C57BL/6J mice frailty, muscle, function, coupled post‐treatment assessments biological age affected molecular pathways. We found that health status, as measured by frailty index, strain, diastolic force, is significantly diminished age, force changing sex‐dependent manner. Conversely, mitigated accumulation was able to partially reverse these declines, evidenced treatment‐induced increases strain fatigue resistance. Despite improvements, did not detect statistically significant changes gene expression or DNA methylation profiles indicative reorganization reduced most ELAM‐treated groups. However, pathway analyses revealed showed pro‐longevity shifts expression, such upregulation genes involved fatty acid metabolism, translation, oxidative phosphorylation, downregulation inflammation. Together, results indicate effective at mitigating signs sarcopenia an aging mouse model, but functional improvements occur independently detectable epigenetic transcriptomic age. Thus, some age‐related may be uncoupled from

Abstract Physical activity and several pharmacological approaches individually combat age-associated conditions extend healthy longevity in model systems. It is tantalizing to extrapolate that combining geroprotector drugs with exercise could beyond any individual treatment. However, the current dogma suggests taking leading on same day as may limit health benefits. Here, we review candidate their interactions highlight salient gaps knowledge need be addressed identify if can have a harmonious relationship exercise.

In the context of increasing number obese individuals, a major problem is represented by obesity and malnutrition in children. This condition mainly ascribable to unbalanced diets characterized high intakes fat sugar. Childhood are not only associated with concurrent pathologies but potentially compromise adult life. Considering strict correlation among systemic metabolism, obesity, skeletal muscle health, we wanted study impact juvenile on muscle. To this aim, 3-week-old C56BL/6 female male mice were fed for 20 weeks high-fat. high-sugar diet, their muscles subjected histological evaluation. MyHCs expression, glycogen content, intramyocellular lipids, mitochondrial activity, capillary density analyzed serial sections obtain metabolic profile. Our observations indicate that high-fat, diet alters profile sex-dependent way induces increase type II fibers, lipid content males, while reducing females. These data highlight response nutrition, calling development specific strategies systematic inclusion subjects basic applied research field.

Temporal muscle thickness (TMT) on cranial CT scans has recently been identified as a prognostic imaging parameter for assessing patient's baseline frailty. Here, we analyzed whether TMT correlates with Traumatic brain injury (TBI) severity and it can be used to predict outcome(s) after TBI. We the radiological clinical data sets of 193 patients TBI who were admitted our institution correlated outcomes stratification TMT. Our analyses showed significant association between high increased risk intracranial hemorrhage (p = 0.0135) but improved mRS at 6 months 0.001) compared low Congruent such findings, lower was associated falls reduced < 0.0001 p 0.0001). High robustly head trauma sequelae also good in patients. These findings consolidate significance an objective marker frailty patients; measurements may ultimately leveraged indicators.
