Those with diabetes mellitus are at high-risk of developing psychiatric disorders, especially mood yet the link between hyperglycemia and altered motivation has not been thoroughly explored. Here, we characterized value-based decision-making behavior a streptozocin-induced diabetic mouse model on Restaurant Row, naturalistic neuroeconomic foraging paradigm capable behaviorally capturing multiple decision systems known to depend dissociable neural circuits. Mice made self-paced choices daily limited time-budget, accepting or rejecting reward offers based cost (delays cued by tone pitch) subjective value (flavors), in closed-economy system tested across months. We found streptozocin-treated mice disproportionately undervalued less-preferred flavors inverted their meal-consumption patterns shifted toward more costly strategy overprioritizing high-value rewards. Abstract Background Increasing evidence implicates astrocytes in stress and depression both rodent models human Major Depressive Disorder (MDD). Despite this, little is known about the transcriptional responses to of within nucleus accumbens (NAc), a key brain reward region, their influence on behavioral outcomes. Methods We used whole cell sorting, RNA-sequencing, bioinformatic analyses investigate NAc astrocyte transcriptome male mice response chronic social defeat (CSDS). Immunohistochemistry was determine stress-induced changes astrocytic CREB NAc. Finally, regulation depression-like behavior investigated using viral-mediated manipulation combination with CSDS. On neuroeconomic foraging tasks, mice, rats, and humans show similar escalations from sunk costs while quitting an ongoing countdown to reward. In a new analysis taken across computationally parallel tasks species laboratories, we find that these behaviors primarily occur on choices are economically inconsistent the subject's other choices, they reflect not only time spent, but also remaining, suggesting change-of-mind re-evaluation processes. The role of altered brain mitochondrial regulation in psychiatric pathologies, including Major Depressive Disorder (MDD), has attracted increasing attention. Aberrant functions were suggested to underlie distinct inter-individual vulnerability stress-related MDD syndrome. In this context, insulin receptor sensitizers (IRSs) that regulate metabolism have become a focus recent research, as their use pre-clinical studies can help elucidate the dynamics disorder and contribute development new antidepressant treatment.
