It is widely accepted that alterations to cyclooxygenase-2 (COX-2) expression and the abundance of its enzymatic product prostaglandin E 2 (PGE ) have key roles in influencing development colorectal cancer. Deregulation COX-2/PGE pathway appears affect tumorigenesis via a number distinct mechanisms: promoting tumour maintenance progression, encouraging metastatic spread, perhaps even participating initiation. Here, we review role signalling highlight ability influence hallmarks cancer—attributes defined by Hanahan Weinberg as being requisite for tumorigenesis. In addition, consider components COX–prostaglandin emerging important regulators tumorigenesis; namely, prostanoid (EP) receptors, 15-hydroxyprostaglandin dehydrogenase transporter. Finally, based on recent findings, propose model cellular adaptation hypoxic microenvironment encompasses interplay between COX-2, hypoxia-inducible factor 1 dynamic switches β-catenin function fine-tune networks meet ever-changing demands tumour.
