In the field of wound treatment, chronic wounds pose a significant burden on medical system, affecting millions patients annually. Current treatment methods often fall short in promoting effective healing, highlighting need for innovative approaches. Electrospinning, technique that has garnered increasing attention recent years, shows promise care due to its unique characteristics and advantages. Recent studies have explored use electrospun nanofibers demonstrating their efficacy cell growth tissue regeneration. Abstract Having a hypoxic microenvironment is common and salient feature of most solid tumors. Hypoxia has profound effect on the biological behavior malignant phenotype cancer cells, mediates effects chemotherapy, radiotherapy, immunotherapy through complex mechanisms, closely associated with poor prognosis in various patients. Accumulating studies have demonstrated that normalization tumor vasculature, nanoparticle carriers biocarriers can effectively increase oxygen concentration microenvironment, improve drug delivery efficacy radiotherapy. They also infiltration innate adaptive anti-tumor immune cells to enhance immunotherapy. Chronic hard-to-heal wounds draw great attention worldwide, as their treatments are limited by infections and hypoxia. Inspired the natural oxygen production capacity of algae competitive advantage beneficial bacteria over other microbes, we presented a living microecological hydrogel (LMH) with functionalized Chlorella Bacillus subtilis encapsulation to realize continuous delivery anti-infections for promoting chronic wound healing. As consisted thermosensitive Pluronic F-127 wet-adhesive polydopamine, LMH could keep liquid at low temperature while quickly solidifying tightly adhering bed. It was demonstrated that optimizing proportion encapsulated microorganism, continuously produce relieve hypoxia support proliferation B. , eliminate colonized pathogenic bacteria. Pathogen infection and tissue injury are universal insults that disrupt homeostasis. Innate immunity senses microbial infections induces cytokines/chemokines to activate resistance mechanisms. Here, we show that, in contrast most pathogen-induced cytokines, interleukin-24 (IL-24) is predominately induced by barrier epithelial progenitors after independent of microbiome or adaptive immunity. Moreover, Il24 ablation mice impedes not only epidermal proliferation re-epithelialization but also capillary fibroblast regeneration within the dermal wound bed. Conversely, ectopic IL-24 induction homeostatic epidermis triggers global epithelial-mesenchymal repair responses. Mechanistically, expression depends upon both IL24-receptor/STAT3 signaling hypoxia-stabilized HIF1α, which converge following trigger autocrine paracrine involving IL-24-mediated receptor metabolic regulation. Thus, parallel innate immune sensing pathogens resolve infections, stem cells sense signals orchestrate repair.

Tissue repair processes maintain proper organ function following mechanical or infection-related damage. In addition to antibacterial properties, mucosal associated invariant T (MAIT) cells express a tissue transcriptomic program and promote skin wound healing when expanded. Herein, we use human-like mouse model of full-thickness excision assess the underlying mechanisms MAIT cell function. Single-cell RNA sequencing analysis suggested that already at steady state. Following excision, promoted keratinocyte proliferation, thereby accelerating healing. Using grafts, parabiosis, adoptive transfer experiments, show migrated into in receptor (TCR)-independent but CXCR6 chemokine receptor-dependent manner. Amphiregulin secreted by Expression was probably independent sustained TCR stimulation. Overall, our study provides mechanistic insights skin.

Whereas the cellular and molecular features of human inflammatory skin diseases are well characterized, their tissue context systemic impact remain poorly understood.
