The chronic airway infections with Pseudomonas aeruginosa are the major co-morbidity in people cystic fibrosis (CF). Within CF lungs, P. persists conducting airways together human mucins as most abundant structural component of its microenvironment. We investigated adhesion 41 serial isolates to mucin preparations from sputa. Mucins and bacteria were retrieved five modulator-naïve patients advanced lung disease. non-CF reference strains showed a strain-specific signature their ovine, porcine bovine submaxillary ranging no or low moderate strong binding. Serial clonal colony morphotypes same sputum sample heterogeneous affinity representatives other clones thus making ‘mucin binding’ one variable intraclonal phenotypic traits known date. Most did not adhere more strongly than plastic surfaces. binders, however, exhibited gradient O -glycans, mammalian mucins.

Abstract Background Mycobacterium avium complex causes chronic and difficult-to-treat infection in vulnerable patient groups, incidence is increasing worldwide. Whole genome sequencing has the potential to reveal new information about how M. persists over time human lung. Methods We analysed genomes of 287 isolates that were sampled longitudinally from 56 patients. Our dataset included 50 newly sequenced a cohort 20 patients Ireland who for up 10 years, we compared these 237 published 2 pre-existing cohorts Europe evaluate strains wider context. Additionally, performed combined analysis across 3 examine changes occurred course infection. Results identified 6 instances where clustered with within 12-SNP threshold, supporting previous observations dominant circulating clones are present internationally. Across cohorts, found communities evolved individual hosts, report acquisition frequent. Importantly, our findings suggest may adapt conditions it faces host, evidence positive selection 12 distinct mycobacterial genes. Notably, multiple virulence-associated genes under selection, including could confer resistance antibiotics host defence mechanisms. Conclusion provides novel insights into within-host evolution highlights potentially important strategies enhance persistence provide targets therapeutic investigation.

In this opinion piece, we consider the meaning of term ‘wild type’ in context microbiology. This is especially pertinent post-genomic era, where have a greater awareness species diversity than ever before. Genomic heterogeneity, vitro evolution/selection pressures, definition ‘the wild’, size and importance pan-genome, gene–gene interactions (epistasis), nature ‘wild-type gene’ are all discussed. We conclude that wild type an outdated even misleading phrase should be gradually phased out.
