Abstract All organisms are exposed to various stressors, which can sometimes lead organismal death, depending on their intensity. While stress-induced death has been observed in many species, the underlying mechanisms remain unclear. In this study, we investigated molecular of fruit fly Drosophila melanogaster . We identified a chymotrypsin-like serine protease Phaedra1 ( Phae1 ) as mediator D. larvae. expression was upregulated by lethal heat stress (40 °C) but not non-lethal (38 °C or lower). The most prominent induction occurred central nervous system (CNS). found neuro-specific knockdown increased survival and reduced neuronal caspase activity following exposure stress, suggesting that transcriptional upregulation CNS is essential for death. next via bioinformatic biochemical analyses transcription factor Zeste (Z) bound enhancer region z loss-of-function impaired CNS, increasing stress. addition, chemical screening rapamycin, inhibitor mechanistic target rapamycin (mTOR), suppressed expression. Postoperative delirium (POD) is generally associated with increased postoperative adverse events. We aimed to investigate whether preoperative quantitative quadriceps muscle ultrasound could predict POD in older patients after gastrointestinal surgery order provide more targeted prevention and treatment measures. prospectively collected data from elderly who underwent elective August December 2023 at a tertiary hospital China. Viral muscle soreness (VMS) is a common feature during acute viral infections, including those caused by enteroviruses, and it substantially diminishes patients’ quality of life. At present, we aim to establish the “brain-muscle” axis explore underlying mechanisms VMS. We initially observed that diminished pain threshold occurred in enterovirus A71 (EV-A71)–infected C57BL/6J AG6 mice. Subsequently, RNA sequencing data showed lipocalin 2 (LCN2) was up-regulated multiple EV-A71, Japanese encephalitis virus, vesicular stomatitis West Nile which all As expected, Lcn2 -deficient C57BL/6 J ( −/− ) mice exhibited greater tolerance, as shown stronger grip force stable motor function after EV-A71 infection. Mechanistically, EV-A71–induced high-mobility group 1 (HMGB1) stimulated astrocyte-derived LCN2 secreted into circulatory system, enhanced glycolysis induced lactate buildup through increasing pyruvate dehydrogenase kinase (PDK1) expression decreasing (PDH) activity. Together, HMGB1/LCN2/PDK1/lactate pathway brain-muscle promoted VMS development.

Post-acute infection syndromes (PAIS), i.e., long-lasting pathologies subsequent to infections that do not properly resolve, have both a common core and broad diversity of manifestations. PAIS include group symptoms (pathological fatigue, cognitive problems, sleep disorders pain) accompanied by large set diverse symptoms. Core additional symptoms, which can persist for years, exhibiting periods relapses remissions, usually start suddenly after an apparently infection. display highly variable clinical features depending on the nature initial pathogen, even larger extent, preexisting individual terrains in are rooted.
