Grass carp reovirus (GCRV) is the most virulent pathogen within genus Aquareovirus, belonging to family Spinareoviridae. GCRV categorized into three genotypes, with type II (GCRV-II) being predominant strain circulating in China. Reoviruses are known replicate and assemble cytoplasmic inclusion bodies termed viroplasms; however, information regarding formation of GCRV-II viroplasms their specific roles virus infection remains largely unknown. In this study, we investigated characteristics during infection. Immunofluorescence confocal microscopy indicate that induces viroplasms, nonstructural protein NS79 key responsible for process. Live-cell imaging fluorescence recovery after photobleaching assays reveal lack liquid-like properties. Transmission electron confirms membranous structures. Notably, demonstrate autophagy autophagosomes utilizes these viroplasm virion assembly. Furthermore, found uses evade host immune system, establishing subclinical persistent also employs nonlytic release viral spread. Collectively, findings highlight distinctive compared those other animal reoviruses, offering valuable insights prevention control virus.IMPORTANCEGrass prevalent Despite reoviruses replication assembly specifics its role were unclear. Our study demonstrates triggers primarily mediated by NS79. structures properties, which significantly different from reoviruses. our research unveils confirm infection, release, results hijacks form complete life cycle. The providing important virus.
