Transcription factors are pivotal regulators in the cellular life process. Activating transcription factor 3 (ATF3), a member of ATF/CREB (cAMP response element-binding protein) family, plays crucial role as cells respond to various stresses and damage. As factor, ATF3 significantly influences signal transduction regulation, orchestrating variety signaling pathways, including apoptosis, ferroptosis, differentiation. In addition, serves an essential link between inflammation, oxidative stress, immune responses. This review summarizes recent advances research on activation its regulating inflammatory responses, cell ferroptosis while exploring dual functions these processes. Additionally, this article discusses diseases related pathogenic microbial infections. Our may be helpful better understand responses disease progression, thus promoting advancements clinical treatments for inflammation stress-related diseases.

Activating Transcription Factor 3 (ATF3) is known for its tumor-suppressive properties in cervical cancer, particularly through role stress response and interactions with human papillomavirus (HPV) oncogenes. This study investigates ATF3's regulatory impact on metastasis-related genes, oxidative stress, DNA damage HPV-positive cancer cells. HeLa Ca Ski cell lines were transfected ATF3-expressing vectors. Western blotting quantitative reverse transcription polymerase chain reaction (RT-qPCR) used to confirm ATF3 overexpression following transfection. ROS assays Comet assessed the of damage, while RT-qPCR was evaluate changes HPV E6/E7, SHARP1, MMP1 gene expression. led elevated levels (p < 0.02), resulting damage. These results demonstrate cytotoxic cells Additionally, significantly decreased expression 0.03), indicating a potential anti-metastatic effect, SHARP1 E6/E7 not altered, selective modulation by ATF3. findings reveal that contributes tumor suppression modulating selectively targeting genes involved metastasis. supports regulating key pathways cells, providing basis further exploration as target therapeutic strategies aimed at improving outcomes cancer.

Cellular integrated stress response (ISR), the mitochondrial unfolded protein (UPRmt), and IFN signaling are associated with viral infections. Activating transcription factor 4 (ATF4) plays a pivotal role in these pathways controls expression of many genes involved redox processes, amino acid metabolism, misfolding, autophagy, apoptosis. The precise ATF4 during infection is unclear depends on cell hosts, agents, models. Furthermore, can be hijacked by pathogens to favor replication. In this review, we summarize ATF4-mediated infections, focusing human immunodeficiency virus 1 (HIV-1). We examine consequences activation for HIV-1 replication reactivation. autophagy apoptosis explored as context programmed deaths contribute depletion CD4 T cells. also participate establishment innate adaptive immunity that essential host control finally discuss putative paralogue, named ATF5, infection. This review underlines at crossroads multiple processes reflecting host–pathogen interactions.
