Marek’s disease virus (MDV), an alphaherpesvirus, causes severe immunosuppression and T cell lymphomas in chickens, known as (MD), economically important poultry primarily controlled by vaccination. Importantly, it also serves a comparative model for studying herpesvirus-induced tumor formation humans. MDV encodes more than 100 genes, most of which have unknown functions. LORF1 is unique to serotype I (MDV-1), lacking homologs other herpesviruses, has not been explored yet. To this end, infectious bacterial artificial chromosome (BAC) harboring the complete genome MDV-1 very virulent strain Md5 was generated, rescued rMd5 maintained biological properties similar parental both vitro vivo . Subsequently, rMd5ΔLORF1, recombinant deficient pLORF1 expression, generated frameshift mutation gene. Chickens infected with rMd5ΔLORF1 exhibited lower mortality rate delayed bursal atrophy those revertant (rMd5-reLORF1). Consistently, viral loads were obviously or rMd5-reLORF1 bursa, but spleen. we found that deficiency impairs replication B cells. Furthermore, showed associated cellular membrane, interacted structural proteins, punctate colocalization tegument capsid proteins cytoplasm. Taken together, study demonstrates first time gene involved MDV-induced formation.
