The tertiary lymphoid structures (TLSs) are positively correlated with the prognosis of many solid tumors, including colorectal cancer. However, their prognostic significance in patients locally advanced rectal cancer (LARC) after neoadjuvant chemotherapy remains unclear. This study aimed to explore correlation between TLS parameters and LARC receiving chemotherapy. retrospective included treated at Harbin Medical University Cancer Hospital from 2012 2021. quantity, area, density TLSs tumor, normal, total tissues surgical specimens were determined. Overall survival (OS) was calculated surgery death any cause. assessed using Kaplan-Meier analysis Cox regression analysis. Multiplex immunofluorescence (mIF) staining used analyze maturity immune composition. 114 patients, whom 46.5% over 60 years old, 70.2% male. tumor region smaller than those normal regions (P < 0.001). A larger area higher (HR = 0.371, P 0.023 for area; HR 0.250, 0.005 density) significantly associated better OS. Moreover, a total-region low carcinoembryonic antigen (CEA) levels 0.028), positive responses therapy 0.001), Subgroup revealed that combining clinicopathologic features such as sex, age, cTNM stage, CEA levels, extramural vascular invasion further stratified prognosis. Additionally, mIF showed high 0.014); mature exhibited greater infiltration CD20⁺ B cells CD21⁺ follicular dendritic compared non-mature TLSs. parameters, particularly density, promising biomarkers undergoing not applicable.

Breast cancer represents the most prevalent form of tumors among females and is characterized by a significant genetic component. The brain frequent site metastasis for breast cancer. Although numerous loci associated with (BCBM) have been identified, critical regulatory genes underlying BCBM remain largely unclear. FinnGen R11 dataset was combined Genotype-Tissue Expression Project (GTEx) Transcriptome-wide Association Study (TWAS). Unified Test Molecular Signatures (UTMOST), Multimarker Analysis Genomic Annotation (MAGMA), Functional Summary-based Imputation (FUSION) were used to identify candidate genes. Summary-data-based mendelian randomization (SMR) co-localization performed further elucidate association between key BCBM. Finally, multiple external cohorts obtained validate findings. In our study, 12 new identified TWAS. Subsequently, both SMR shown that CAPS8 only expressed in tissues including frontal cortex cerebellar hemispheres Potential regulation CASP8 could occur findings ultimately validated clinical cohorts. Abstract Brain metastases (BrM) are the most common cancers in brain and linked to poor prognosis. Given high incidence often limited treatment options, understanding complexity of BrM tumor microenvironment is crucial for development novel therapeutic strategies. We performed transcriptome-wide gene expression profiling combined with spatial immune cell characterize 95 patients from different primary tumors. found that lung carcinoma malignant melanoma showed overall higher infiltration as compared breast carcinoma. RNA sequencing-based deconvolution revealed signatures indicative tertiary lymphoid structures (TLS) subsets BrM, mostly cancer melanoma. This finding was corroborated by multiplex immunofluorescence staining cells tissue sections. Detection TLS more treatment-naïve associated prolonged survival after diagnosis patients. Our findings highlight cellular diversity types suggest a role formation patient outcome.

Esophageal cancer (EC), especially esophageal squamous cell carcinoma (ESCC), which is the most common subtype in China, one of aggressive gastrointestinal malignancies. Traditional treatments like surgery, radiotherapy, and chemotherapy have limited success, but study tertiary lymphoid structure (TLS) has opened new avenues for immunotherapy. TLS an ectopic immune cluster, including B cells, T dendritic cells (DCs), that forms chronic inflammation such as tumors. often found at tumor invasion margins linked to better prognosis EC patients, with higher density maturation associated prolonged survival. can also serve a biomarker assess immunotherapy outcomes. Further exploration molecular mechanisms could innovate treatments, by inducing formation improve therapy. This review summarizes characteristics EC, clinical findings, limitations directions current research, offering insights future strategies.
