The brain is an exceptionally lipid-rich organ with a very complex lipid composition. Lipids are central in several neuronal processes, including membrane formation and fusion, myelin packing, lipid-mediated signal transmission. Lipid diversity associated the evolution of higher cognitive abilities primates, affected by activity, instrumental for synaptic plasticity, illustrating that lipids not static components membranes. Several lines evidence suggest composition synapses unique distinct from other subcompartments. Here, we delve into nascent field synaptoneurolipidomics, offering overview current knowledge on junctions technological advances will allow us to study impact function.

Central nervous system (CNS) diseases, such as brain tumors, Alzheimer’s disease, and Parkinson’s significantly impact patients’ quality of life impose substantial economic burdens on society. The blood–brain barrier (BBB) limits the effective delivery most therapeutic drugs, especially natural products, despite their potential effects. Trojan Horse strategy, using nanotechnology to disguise drugs “cargo”, enables them bypass BBB, enhancing targeting efficacy. This review explores applications products in treatment CNS discusses challenges posed by analyzes advantages limitations strategy. Despite existing technical challenges, future research is expected enhance application integrating nanotechnology, improving mechanisms, optimizing characteristics.

Serotonin is distinct among synaptic neurotransmitters because it amphipathic and released from vesicles at concentrations superior to its water solubility limit (270 mM in for a of 110 mM). Hence, serotonin mostly aggregated the cleft, due extensive aromatic stacking. This important characteristic has received scant attention, as most representations serotonergic synapse take warranted that molecules are present monomers after vesicle exocytosis. Using combination silico physicochemical approaches new experimental device mimicking conditions, we show aggregates efficiently dissolved by gangliosides (especially GM1) postsynaptic membranes. initial interaction, driven electrostatic forces, attracts insoluble resolves micelles into monomers. also interacts with cholesterol via set CH-π van der Waals interactions. Thus, act together functional serotonin-collecting funnel on brain cell Based this unique mode interaction membranes, propose model transmission takes account post-exocytosis solubilizing effect aggregates.
