Systemic lupus erythematosus (SLE) is a chronic, systemic autoimmune disease with wide range of clinical expressions. The kidney often affected, usually within 5 years the onset SLE, and nephropathy (LN) carries high risk for increased morbidity. heterogeneity accompanied by complex disturbances affecting immune system inflammation tissue damage due to loss tolerance nuclear antigens deposition complexes in tissues. Several studies have reported that human there an important role Type-I-interferons (INF) suggested upregulation INF-inducible genes observed serial gene expression microarray studies. This review aims describe transduction pathways Type-I-interferons, particular INFα, its immune-regulatory function pathogenesis SLE and, particular, LN. In addition, recent novelties concerning biologic therapy LN will be discussed.

Women have stronger immune responses to infections and vaccination than men. Paradoxically, the response comes at a steep price, which is high incidence of autoimmune diseases in women. The reasons why women immunity higher autoimmunity are not clear. Besides gender, sex hormones contribute development activity system, accounting for differences gender-related responses. Both innate adaptive systems bear receptors respond hormonal cues. This review focuses on role particularly estrogen, response, health, disease with an emphasis systemic lupus erythematosus.

Many studies indicate that microRNAs (miRNAs) could be potential biomarkers for various diseases. The purpose of this study was to investigate the clinical value serum exosomal miRNAs in systemic lupus erythematosus (SLE).Serum exosomes were isolated from 38 patients with SLE and 18 healthy controls (HCs). expression miR-21, miR-146a miR-155 within examined by reverse transcription-quantitative polymerase chain reaction (RT-qPCR). Using receiver operating characteristic (ROC) curves, we evaluated diagnostic miRNAs.Exosomal miR-21 upregulated (p<0.01), whereas (p<0.05) downregulated SLE, compared HCs. (p<0.01) higher nephritis (LN) than those without LN (non-LN). analysis ROC curves revealed showed a LN. Furthermore, (R=0.44, p<0.05) (R=0.33, positively correlated proteinuria. negatively associated anti-SSA/Ro antibodies (R=-0.38, p<0.05), anti-dsDNA (R=-0.39, p<0.05).These findings suggested levels may serve as diagnosis

Objective: To identify microRNAs (miRNAs) differentially expressed in plasma exosomes collected women diagnosed with preeclampsia compared uncomplicated pregnancies.Materials and methods: Exosomes were purified from samples obtained at each trimester four subsequently five matched healthy controls. RNA was the exosomes, expression of 368 miRNAs profiled using A-Set TaqMan low density array (TLDA).Results: One-third are not exosomes. Further, those that tend to be evolutionarily younger have a significantly different mature sequence signature than do Among total eight (miR-134, miR-196b, miR-302c, miR-346, miR-376c, miR-486-3p, miR-590-5p, miR-618) found display statistically significant differential between who developed as did not. Moreover, half these miR-590-5p) displayed first trimester. Conclusions: Not all Those older A few exosome-expressed patterns having an and, among these, several appear trimester. These potential early markers risk.

Systemic lupus erythematosus (SLE) is a serious autoimmune disease with variety of organ manifestations. The most dreadful one, affecting the majority SLE patients, kidney manifestation – nephritis (LN). Dendritic cells (DC) are believed to be one culprits immune dysregulation in LN. Flow cytometry analysis was applied identify frequency and activity peripheral blood DCs subpopulations: myeloid plasmacytoid, LN patients. Magnetically isolated mDCs pDCs were subjected molecular genes expression, evaluation global DNA methylation histone H3 methylation. We observed distinctive features associated stages Lower numbers patients severe LN, while increased costimulatory potential connected early, mild stage this disease. IRF1 transcript upregulation specific for from total exceptional amounts mRNA detected hypermethylation seemed characteristic whereas decrease H3K4me3 H3K27me3 marks significant early These findings present dendritic cell alterations that may reflect renal involvement SLE, laying foundations new strategy diagnosis monitoring omitting invasive biopsies.

Systemic lupus erythematosus (SLE) is an archetype of systemic autoimmune disease, characterized by the presence diverse autoantibodies and chronic inflammation. There are multiple factors involved in pathogenesis, including genetic/epigenetic predisposition, sexual hormone imbalance, environmental stimulants, mental/psychological stresses, undefined events. Recently, many authors noted that “inflammaging”, consisting immunosenescence inflammation, a common feature aging people patients with SLE. It conceivable oxidative stresses originating from mitochondrial dysfunction, defective bioenergetics, abnormal immunometabolism, premature telomere erosion may accelerate immune cell senescence The dysfunctions SLE have been extensively investigated recent years. molecular basis normoglycemic metabolic syndrome has found to be relevant production advanced glycosylated nitrosative end products. Besides, immunosenescence, autoimmunity, endothelial damage, decreased tissue regeneration could results Herein, cellular bases inflammaging cardiovascular complications will reviewed aspects dysfunctions, bioenergetics/immunometabolism, telomere/telomerase disequilibrium.

Systemic lupus erythematosus (SLE) is a prototype of systemic autoimmune disease involving almost every organ. Polygenic predisposition and complicated epigenetic regulations are the upstream factors to elicit its development. Mitochondrial dysfunction-provoked oxidative stress may also play crucial role in it. Classical gene expression include DNA methylation/acetylation histone modification. Recent investigations have revealed that intracellular extracellular (exosomal) noncoding RNAs (ncRNAs), including microRNAs (miRs), long (lncRNAs), key molecules for post-transcriptional regulation messenger (m)RNA expression. Oxidative nitrosative stresses originating from mitochondrial dysfunctions could become pathological biosignatures increased cell apoptosis/necrosis, nonhyperglycemic metabolic syndrome, multiple neoantigen formation, immune dysregulation patients with SLE. Recently, many authors noted cross-talk between ncRNAs can trigger perpetuate reactions Intracellular interactions miR lncRNAs as well exosomal ncRNA communication fro remote cells/tissues via plasma or other body fluids occur body. The urinary now represent nephritis. Herein, we'll briefly review discuss excessive oxidative/nitrosative induced by dysfunction tissues/cells ncRNAs, prospect antioxidant therapy

MicroRNAs (miRs) are non-coding small RNAs that act as epigenetic modulators to regulate the protein levels of target mRNAs without modifying genetic sequences. The role miRs in pathogenesis lupus nephritis (LN) is increasingly recognized and highly complex. Altered different observed blood, urine kidney tissues murine LN models patients. Accumulating evidence suggests these can modulate immune cells various key inflammatory pathways, their perturbations contribute aberrant response LN. dysregulation resident renal urinary exosomes also lead abnormal cell proliferation, inflammation fibrosis While may hold promise clinical applications patients, there still many potential limitations safety concerns for use. Further studies worthwhile examine utility diagnosis, disease activity monitoring, prognostication treatment
