Oxidative stress is a silent participant in all chronic progressive pathology and degenerative disorders. Neurodegeneration cognitive impairment are caused by increased imbalanced oxidative down-regulated natural anti-oxidant combat mechanisms. Cancer the end result of cell malfunction activation tumour-specific genes, which regulated specific geneomolecular pathways. Acute stroke decrease body's combating defence stress, results from an imbalance between antioxidative activities cells, has been linked to aetiology number diseases, including cancer stroke. The harmful effects ROS their role lesion progression after ischaemic undeniable; however, therapeutic usefulness anti-oxidants this scenario remains debatable. In review, we have summarised potent molecular mechanisms play vital through various pathways associated advanced targeted therapeutics like pathologies.

Aging is a normal, inevitable, irreversible, and progressive process which driven by internal external factors. Oxidative stress, that the imbalance between prooxidant antioxidant molecules favoring first, plays key role in pathophysiology of aging comprises one molecular mechanisms underlying age-related diseases. However, oxidative stress theory has not been successfully proven all animal models studying lifespan, meaning altering stress/antioxidant defense systems did always lead to prolonged as expected. On other hand, pathological phenotypes showed well-correlated relationship with levels molecules. Therefore, it seems more complicated than once believed this might be affected environment each organism. Environmental factors such UV radiation, air pollution, an unbalanced diet, have also implicated seem initiate rapidly even at younger ages. The purpose review elucidate physiology effect certain environmental initiating sustaining process. Understanding will contribute development strategies postpone phenomenon. In addition, recent studies investigating ways alter order prevent presented.Careful exposure harmful use supplements could potentially affect biological processes driving slow down Maybe lifespan achieved strategy alone, but longer healthspan favorable target.

Prolonged elevated oxidative stress (OS) possesses negative effect on cell structure and functioning, is associated with the development of numerous disorders. Naturally occurred anti-oxidant compounds reduce in living organisms. In this review, antioxidant properties β-carotene, tocopherols ascorbic acid are presented based vitro, vivo populational studies. Firstly, environmental factors contributing to OS occurrence intracellular sources Reactive Oxygen Species (ROS) generation, as well ROS-mediated cellular degradation, introduced. Secondly, enzymatic non-enzymatic mechanism defence against development, presented. Furthermore, ROS-preventing mechanisms effectiveness anti-oxidants summarized, studies where different ROS-generating (oxidizing) agents used. Oxidative biomarkers, indicators level prevention by supplementation, a focus methods (spectrophotometric, fluorometric, chromatographic, immuno-enzymatic) their detection. Finally, application Raman spectroscopy imaging tool for monitoring (β-carotene, acid) metabolism, proposed. Literature data gathered suggest that possess potential mitigate various biological systems. Moreover, can be valuable technique study molecules

The excessive production of various reactive oxidant species over endogenous antioxidant defense mechanisms leads to the development a state oxidative stress, with serious biological consequences. consequences stress depend on balance between generation and include damage biomolecules, disruption signal transduction, mutation, cell apoptosis. Accumulating evidence suggests that is involved in physiopathology debilitating illnesses associated chronic inflammation, including cardiovascular diseases, diabetes, cancer, or neurodegenerative processes, need continuous pharmacological treatment. Oxidative inflammation are tightly linked pathophysiological one which can be simply promoted by another. Although, many trials have been unsuccessful (some showed either no effect even harmful effects) human patients as preventive curative measure, targeting remains an interesting therapeutic approach for new agents design novel anti-inflammatory drugs reliable safety profile. In this regard, several natural compounds were explored potential options treatment inflammatory diseases. Several metalloenzymes, such superoxide dismutase, catalase, glutathione peroxidase, among essential enzymes maintain low nanomolar physiological concentrations (O 2 •−) hydrogen peroxide (H O ), major redox signaling molecules, thus play important roles alteration homeostasis. These become striking source motivation catalytic enhance action these under pathological conditions related inflammation. This review focused representatives synthetic antioxidants drug candidates
