Abstract Background Gastrointestinal stromal tumor (GIST) is the most common human sarcoma with over 5000 new patients diagnosed in USA each year. The originates from interstitial cells of Cajal and forms an intramural lesion commonly stomach or small intestine. gut microbiome has been linked to other gastrointestinal cancers a recent paper purported that GISTs contain substantial intratumoral bacteria. purpose this study further evaluate presence bacteria GISTs. Patients Methods We collected 25 samples varying size location 24 under sterile conditions operating room immediately following surgical resection. 16S quantitative polymerase chain reaction (qPCR) ribosomal RNA (rRNA) gene amplicon sequencing were performed bacterial species present tumor. Retrospective chart review was determine characteristics, including size, location, imatinib exposure, mucosal involvement. Results In 23 samples, there fewer than 100 copy numbers rRNA per uL, indicating absence significant load. remaining two one gastric duodenal tumor, revealed normal intestinal These tumors, along three others, had disruption lining. Conclusions generally lack bacteria, except some cases when disrupts mucosa.

ABSTRACT Background The mycobiome in the tumor microenvironment of non‐smokers with early‐stage lung adenocarcinoma (ES‐LUAD) has been minimally investigated. Methods In this study, we conducted ultra‐deep metagenomic and transcriptomic sequencing on 128 samples collected from 46 nonsmoking ES‐LUAD patients 41 healthy controls (HC), aiming to characterize tumor‐resident its interactions host. Results results revealed that exhibited fungal dysbiosis characterized by reduced species diversity significant imbalances specific abundances. Concurrently, microbial functional analysis alterations associated genes such as ribosomal proteins histones. We observed correlations between Yarrowia lipolytica , Saccharomyces paradoxus tumor‐infiltrating immune cells (TIICs), identified a strong association (|rho| > 0.7) S. 14 transcription factors. A signature three prognostic (GRIA1, CDO1, FHL1) closely was they suggest interaction host may contribute prolonged overall survival (OS). Finally, predictive model based six fungi demonstrated decent classification performance distinguishing cases HCs (AUC = 0.724). Conclusions Our study demonstrates transcriptome within tumors help elucidate pathogenic mechanisms ES‐LUAD. Fungi, potential tool, can be used an additional resource for accurately detecting discriminating individuals

Abstract Research has shown that the microbiome can influence how immune system responds to melanoma cells, affecting course of disease and outcome therapy. Here, we used metagenomic approach flow cytometry analyses blood cells discover correlations between gut fungi metastatic patients enrolled in anti-PD-1 therapy lymphocytes their blood. We analyzed patterns associations before first administration (BT, n = 61) third month (T3, 37), allowing us track changes during treatment. To understand possible impact on efficacy therapy, clinical beneficiaries (CB, 37) non-beneficiaries (NB, 24), as well responders (R, 28) non-responders (NR, 33). Patients with LDH < 338 units/L, overall survival (OS) > 12, CB, R, had lower levels Shannon diversity ( p 0.02, 0.05, 0.03, respectively). found correlation pattern intestinal was specific type response, positive or negative. When comparing CB NB groups, opposite directions were detected for C. albicans , suggesting a response-specific reaction. For M. restricta exhibited set different types lymphocytes, prevalent correlations, robust response group. This result extends our former research, where associated an increased risk progression poorer

Overwhelming evidence suggests that the gut microbiota modulates tumor response to immune checkpoint inhibitors (ICIs). The probiotic Bacteroides fragilis (BF839) was extensively used in China improve dysbiosis-related symptoms. We hypothesized BF839 could enhance sensitivity ICIs. In preclinical studies, mice received orally, PD-1 intraperitoneal injection, or a combination therapy of two agents. antitumor effect investigated by assessing growth and microenvironment. Mice fecal samples were collected for 16S rRNA sequencing. Fresh RNA data 29 patients with advanced solid who adjuvant retrospectively evaluated. primary endpoint overall survival (OS). Among tumors undergoing ICIs chemotherapy, long-term treatment group had longer OS (p = 0.0101) than short-term group. we found monotherapy anti-PD-1 antibody significantly inhibit growth. Interestingly, worked synergistically induced regression, mediated increased CD8+T cell infiltration. Mechanistically, suppression regulated cGAS-STING pathway. sequencing results showed diversity. Overall, our suggest enhanced through signaling. future, application regulate may be new strategy efficacy

Background Recent findings highlight the significant impact of intestinal fungi on complex makeup gut microbiota and human health, challenging past oversights. However, a lack thorough systematic quantitative analyses remains. This study aims to address this gap by thoroughly examining current research fungi. Through analyzing developments unique features in area, our goal is foster deeper understanding identify future pathways. Methods We performed an extensive bibliometric analysis documents from 2000 2023, sourced Web Science Core Collection (WoSCC). Utilizing advanced visualization tools such as VOSviewer, CiteSpace, Bibliometrix R, we meticulously examined illustrated data scientific landscapes networks. Results A total 1434 papers were analyzed, revealing substantial increase publication volume over two decades, particularly 2020. Contributions came 67 countries, 2178 institutions, 8,479 authors. China led output with 468 articles, followed University California 84 ZHANG F most prolific author 17 articles. Emerging areas “Fungal-Bacteria Interactions,” “Gut Fungus Gut-Brain Axis,” Immunity” are expected attract growing interest future. Conclusion offers overview scholarly efforts concerning fungi, highlighting predominant landscape field. These insights can assist scholars identifying appropriate avenues, forming collaborative relationships, enhancing key themes emerging areas, thereby stimulating endeavors.
