Senescence is a crucial player in several metabolic disorders and chronic inflammatory diseases. Recent data prove the involvement of hepatocyte senescence development NAFLD (non-alcoholic fatty liver disease). As main energy ROS (reactive oxygen species) producing organelle, mitochondria play central role accelerated diseases development. In this review, we focus on regulation mitochondrial Ca2+ homeostasis, NAD+/NADH ratio, UPRmt (mitochondrial unfolded protein response), phospholipids acid oxidation hepatic senescence, lifespan disease susceptibility. Additionally, nuclear mutations lifespan-modulation discussed. While DNA SNPs (single nucleotide polymorphisms) can be used as effective diagnostic markers targets for treatments, advanced age should considered an independent risk factor

Sleep is a modifiable behavior that can be targeted in interventions aimed at promoting healthy aging. This study aims to (i) identify the sleep duration trend US adults; (ii) investigate relationship between and phenotypic age; (iii) explore role of exercise this relationship. Phenotypic age as novel index was calculated according biomarkers collected from adults based on National Health Nutrition Examination Survey (NHANES). information self-reported by participants discerned through individual interviews. The principal analytical method employed weighted multivariable linear regression modeling, which accommodated for complex multi-stage sampling design. potential non-linear explored using restricted cubic spline (RCS) model. Furthermore, subgroup analyses evaluated effects sociodemographic lifestyle factors primary outcomes. A total 13,569 were finally included in, thereby resulting population 78,880,615. An examination temporal trends revealed declining proportion individuals with insufficient markedly deficient time since 2015-2016 cycle. Taken normal group reference, extreme short [β (95% CI) 0.582 (0.018, 1.146), p = 0.044] long 0.694 (0.186, 1.203), 0.010] both positively associated fully adjusted According dose-response age, benefit regular activity, whereas more tended have higher age. There an inverted U-shaped durations represents important step forward our understanding By shedding light topic providing practical recommendations habits, researchers help live longer, healthier, fulfilling lives.

Cellular senescence is understood to be a biological process that defined as irreversible growth arrest and was originally recognized tumor-suppressive mechanism prevents further propagation of damaged cells. More recently, cellular has been shown have dual role in prevention tumor promotion. Senescent cells carry senescence-associated secretory phenotype (SASP), which altered by factors including pro-inflammatory cytokines, chemokines, other proteases, leading the alteration tissue microenvironment. Though would eventually halt cancerous potential cells, SASP contributes environment promoting inflammation, matrix remodeling, cell invasion. The paradox prevention/promotion particularly relevant bone niche microenvironment, where longer-lasting, chronic inflammation promotes formation. Insights into mechanistic understanding provide basis for targeted therapies, such senolytics, aim eliminate senescent or inhibitors, tumor-promoting effects senescence. These therapeutic interventions offer significant clinical implications treating cancer healthy aging.

The liver is a unique organ with an abundant regenerative capacity. Therefore, partial hepatectomy (PHx) or transplantation (PLTx) can be safely performed. Liver regeneration involves complex network of numerous hepatotropic factors, cytokines, pathways, and transcriptional factors. Compared after viral- drug-induced injury, that post-PHx -PLTx has several distinct features, such as hemodynamic changes in portal venous flow pressure, tissue ischemia/hypoxia, hemostasis/platelet activation. Although some these also occur during they are more abrupt drastic following PHx PLTx, thus the main trigger driving force regeneration. In this review, we first provide overview molecular biology -PLTx. Additionally, phospho (p)-RPS6 has been recognized as surrogate marker for activated PI3K/AKT/mTORC1 pathway, which occurs many cancer types. However, downstream mechanisms regulated by RPS6 or p-RPS remains elusive, therapeutic implication underappreciated despite an approximately half century history research on this protein.
