Transplantation of corneal tissue is the standard treatment for irreversible endothelium decompensation by replacing malfunctioning endothelial cells and Descemet's membrane. However, this surgery depends on limited donor supply. Thus, developing suitable alternatives grafting material through a engineering approach needed to enable transplantation cultured restore normal function. Here, we proposed using plastic compressed collagen 3D matrix called Real Architecture For Tissues (RAFT) as scaffold cell culture. The porcine cornea (PCECs) were seeded RAFT construct tissue-engineered endothelium. Then, cell-seeded-RAFT graft was transplanted onto human maintained in an ex vivo organ culture model. results showed that PCECs formed high-density monolayer expressing markers, ZO-1, Na/K ATPase N-cadherin. More importantly, cell-seeded successfully restored function, drawing thickness endothelium-wounded back two weeks
